【摘要】：目的:探討tau蛋白磷酸化在阿爾茨海默病不同中醫(yī)證型中的變化特征。方法:40只雄性SD大鼠隨機分為假手術(shù)組、疾病模型組、腎精虧虛證組、痰濁阻竅證組。除假手術(shù)組及疾病模型組外,其余組均予長期皮下注射D-半乳糖聯(lián)合雙側(cè)海馬CA1區(qū)定位注射凝聚態(tài)Aβ25-35構(gòu)建病證結(jié)合腎精虧虛證大鼠模型,在此基礎(chǔ)上痰濁阻竅證組給予高脂飼料構(gòu)建痰濁阻竅證模型。采用Morris水迷宮、尼氏染色、Western blot等方法,觀察各組大鼠學(xué)習(xí)記憶、海馬神經(jīng)元凋亡及tau蛋白過度磷酸化特異性位點的改變。結(jié)果:與假手術(shù)組比較,疾病模型組、腎精虧虛證組、痰濁阻竅證組大鼠逃避潛伏期未見明顯縮短,目標(biāo)象限停留時間明顯減少(P0.01)、穿越平臺的次數(shù)也明顯減少(P0.01)。與假手術(shù)組比較,疾病模型組、腎精虧虛證組、痰濁阻竅證組尼氏染色均出現(xiàn)不同程度的神經(jīng)元凋亡(P0.05,P0.01);與疾病模型組比較,痰濁阻竅證組凋亡最明顯(P0.01)。與疾病模型組、腎精虧虛證組比較,痰濁阻竅證組大鼠海馬組織tau蛋白Thr181位點的磷酸化均顯著增加(P0.01)。結(jié)論:tau蛋白Thr181位點的過度磷酸化可能是AD痰濁阻竅證的特征性微觀病理改變。
[Abstract]:Objective: to investigate the changes of tau protein phosphorylation in different TCM syndromes of Alzheimer's disease. Methods: forty male SD rats were randomly divided into sham operation group, disease model group and kidney essence deficiency syndrome group. Except sham-operation group and disease model group, all the other groups were given long-term subcutaneous injection of D-galactose and localization injection of condensed matter A 尾 25-35 in bilateral hippocampal CA1 region to construct the rat model of disease and syndrome combined with deficiency of kidney essence. On this basis, the model of phlegm turbid obstruction of orifices was established in the phlegm turbid obstructive orifices syndrome group with high fat feed. The learning and memory of rats in each group were observed by Morris water maze and Nissl staining Western blot. Results: compared with the sham-operated group, the escape latency of rats in the disease model group, kidney essence deficiency syndrome group and phlegm turbid obstruction syndrome group was not significantly shortened. Compared with sham operation group, the disease model group, kidney essence deficiency syndrome group, disease model group, kidney essence deficiency syndrome group, disease model group, kidney essence deficiency syndrome group, disease model group, kidney essence deficiency syndrome group, disease model group, kidney essence deficiency syndrome group, disease model group, kidney essence deficiency syndrome group, Compared with the disease model group, the apoptosis of phlegm turbid obstruction syndrome group was the most obvious (P 0.01), and compared with the disease model group and kidney essence deficiency syndrome group, the apoptosis of phlegm turbid obstruction orifices syndrome group was more obvious than that of the disease model group, and compared with the disease model group and the kidney essence deficiency syndrome group. The phosphorylation of Thr181 site of tau protein in hippocampus of rats with phlegm turbid obstruction of orifices was significantly increased (P 0.01). Conclusion the excessive phosphorylation of Thr181 site of 10 tau protein may be the characteristic microscopic pathological change of AD syndrome of phlegm turbid obstruction of orifices.
