超聲生物顯微鏡對溫經(jīng)通絡(luò)法干預(yù)膠原誘導(dǎo)性關(guān)節(jié)炎大鼠滑膜增生的研究

【摘要】:目的:以超聲生物顯微鏡(UBM)觀察溫經(jīng)通絡(luò)法干預(yù)膠原誘導(dǎo)性關(guān)節(jié)炎(CIA)大鼠滑膜增生情況。方法:40只SPF級雌性Wistar大鼠,隨機分為正常組、模型組、對照組和中藥組,每組10只。正常組常規(guī)飼養(yǎng),其余各組復(fù)制膠原誘導(dǎo)性關(guān)節(jié)炎(CIA)大鼠模型后,分別以蒸餾水、雷公藤多苷片和加味芍甘附子湯干預(yù)4周,觀察大鼠關(guān)節(jié)炎癥評分(AI)和雙足排水體積,超聲生物顯微鏡觀察治療后各組大鼠跖趾關(guān)節(jié)病變情況,并與組織病理學(xué)結(jié)果進行對比。結(jié)果:造模后14~21 d,除正常組外各組AI值和雙足排水體積均顯著升高,給藥后各組腫脹持續(xù)減輕,AI值降低。造模后42 d,中藥組大鼠右足排水體積顯著低于對照組(P0.05),同時期各組間AI和雙足排水體積比較差異無統(tǒng)計學(xué)意義(P0.05);UBM平掃正常組可見骨皮質(zhì)為連續(xù)的、致密強回聲帶,滑膜無顯著增厚,厚度低于0.1 mm,其余3組關(guān)節(jié)間隙內(nèi)均可見不同程度中等回聲信號,其中模型組骨面毛糙,邊緣欠整齊最明顯,與病理學(xué)結(jié)果相符;滑膜病理評分與滑膜厚度呈正相關(guān)(r=0.637,P=0.002),與AI評分無顯著相關(guān)性(r=0.192,P=0.344)。結(jié)論:UBM可用于檢測RA動物模型早期病理改變,對療效觀察和預(yù)后評估具有較好的潛在價值。
[Abstract]:Objective: to observe the synovium proliferation of collagen-induced arthritis (CIA) rats by ultrasound biomicroscopy (UBMM). Methods: 40 SPF female Wistar rats were randomly divided into normal group, model group, control group and traditional Chinese medicine group. Ten rats in each group were fed with the normal control group, and the other groups were treated with distilled water, tripterygium wilfordii polyglycoside tablets and modified Paeoniao Gan Fuzi decoction for 4 weeks after the model of collagen-induced arthritis (CIA) was established in each group. The arthritis score of rats (AII) and the drainage volume of both feet were observed. The pathological changes of metatarsophalangeal joint were observed by ultrasound biomicroscopy. The results were compared with those of histopathology. Results: the AI value and biped drainage volume increased significantly in each group except the normal group at day 14 and 21 after modeling. The volume of draining of right foot in the Chinese medicine group was significantly lower than that of the control group at 42 days after administration of the drug, and there was no significant difference in AI and the volume of drainage between the two groups at the same time. We can see that the bone cortex is continuous. The thickness of synovium was less than 0.1 mm and the thickness of synovium was less than 0.1 mm. The other three groups showed moderate echo signal in the space of joint, in which the bone surface of model group was rough and the edge was not neat, which was consistent with the pathological results. There was a positive correlation between synovial pathological score and synovial thickness. There was no significant correlation between synovial pathological score and AI score. Conclusion: the early pathological changes of RA animal model can be detected by WUBM, which has a good potential value for curative effect observation and prognosis evaluation.
