按摩聯(lián)合針刺療法對大鼠運動型骨骼肌損傷的保護(hù)作用

【摘要】:目的探討按摩聯(lián)合針刺療法對大鼠運動型骨骼肌損傷肱三頭肌超微結(jié)構(gòu)及磷酸肌酸激酶的影響及對運動型骨骼肌損傷的保護(hù)作用。方法將130只SD大鼠隨機分為非運動組(安靜對照組)和運動組,運動組按照干預(yù)方式不同分為運動對照組、按摩組、針刺組、聯(lián)合組,每一運動組又按照樣品采集時間不同分為以下亞組,包括即刻組、24 h組、48 h組。共計13組,每組10只大鼠。運動組在大鼠跑臺上進(jìn)行持續(xù)性下坡跑,做力竭離心運動。力竭運動后對照組不做任何處理,按摩組每日二次給予肱三頭肌的揉捏推壓和牽引按摩5 min。針刺組給予每日一次毫針斜刺肱三頭肌,留針5 min。聯(lián)合組在給予按摩治療同時給予針刺療法。實驗方案完成后在運動后即刻、24 h、48 h取大鼠血清行磷酸肌酸激酶檢測。取肱三頭肌組織在鏡下觀察超微結(jié)構(gòu)的變化,主要為細(xì)胞結(jié)構(gòu)的異常與Z線的異常。結(jié)果力竭離心運動后大鼠肱三頭肌超微結(jié)構(gòu)發(fā)生改變,包括細(xì)胞結(jié)構(gòu)異常(肌纖維排列紊亂,細(xì)胞膜模糊、溶解,線粒體腫脹、空泡樣改變)和Z線異常(包括Z線流、Z線模糊、Z線扭曲和Z線消失)。這些超微結(jié)構(gòu)異常改變在運動后24 h最明顯,48 h有所恢復(fù)。而磷酸肌酸激酶在運動后即刻開始顯著升高,48 h有所恢復(fù),但仍高于安靜對照組。各治療組超微結(jié)構(gòu)損害在24 h和48 h均較運動對照組減輕,聯(lián)合治療組損傷最輕;磷酸肌酸激酶值在24 h和48 h較運動對照組降低。聯(lián)合治療組磷酸肌酸激酶值最低。結(jié)論 (1)力竭離心運動可引起骨骼肌超微結(jié)構(gòu)損害和磷酸肌酸激酶升高。(2)磷酸肌酸激酶更能早期反映骨骼肌的損傷。(3)按摩聯(lián)合針刺療法可以減輕骨骼肌超微結(jié)構(gòu)的損害和降低磷酸肌酸激酶,說明兩者聯(lián)合對運動型骨骼肌損傷具有保護(hù)作用。
[Abstract]:Objective to investigate the effects of massage combined with acupuncture therapy on the ultrastructure of triceps brachii muscle and the protective effect of phosphocreatine kinase on exercise skeletal muscle injury in rats. Methods 130 Sprague-Dawley rats were randomly divided into two groups. Exercise group (quiet control group) and exercise group, The exercise group was divided into exercise control group, massage group, acupuncture group and combined group according to the intervention methods. Each exercise group was divided into the following subgroups according to sample collection time, including immediate group, 24 h group, 48 h group, total 13 groups. There were 10 rats in each group. The exercise group performed continuous downhill running and exhaustive centrifugation on the platform of the rats. The control group did not do any treatment after exhaustive exercise. The massage group was given the kneading and pressing of triceps brachii twice a day and the traction massage for 5 mins, while the acupuncture group was given the triceps brachii muscle once a day by oblique needling. The rats in the combined group were treated with massage and acupuncture at the same time. The serum samples of rats were taken immediately after exercise for 24 hours and 48 hours after exercise to detect creatine phosphokinase. The changes of ultrastructure of triceps brachii muscle tissue were observed under microscope. Results after exhaustive centrifugation, the ultrastructure of triceps brachii muscle changed, including abnormal cell structure (disordered arrangement of muscle fibers, blurred cell membrane, dissolution of mitochondria, swelling of mitochondria). Vacuolar changes) and Z-line abnormalities (including Z-line flow, fuzzy Z-line distortion and Z-line disappearance). These ultrastructural changes were most pronounced at 24 h after exercise and recovered at 48 h after exercise, while creatine phosphokinase recovered immediately after exercise. It began to increase significantly at 48 h, and recovered after 48 h. The ultrastructural damage in each treatment group was lighter than that in the exercise control group at 24 h and 48 h, and the damage in the combined treatment group was the least. The value of phosphocreatine kinase at 24 h and 48 h was lower than that in the exercise control group, and the value of phosphocreatine kinase was the lowest in the combined treatment group. Conclusion the exhaustive centrifugal exercise can cause ultrastructural damage of skeletal muscle and increase of creatine phosphokinase. Creatine kinase can reflect skeletal muscle injury in early stage. Massage combined with acupuncture therapy can reduce the damage of skeletal muscle ultrastructure and reduce creatine phosphokinase. These results suggest that the combined therapy has protective effect on the injury of sports skeletal muscle.
