【摘要】：目的:研究針刺"天樞穴"對(duì)DSS誘導(dǎo)結(jié)腸炎(IBD)的治療作用及其對(duì)核因子-κB(NF-κB)信號(hào)通路的影響。方法:將30只C57BL/6小鼠隨機(jī)分為空白組、模型組和天樞穴組,模型組和天樞穴組分別給予3%DSS水溶液造模,天樞穴組每日給予20min針刺干預(yù),連續(xù)7d。記錄每日DAI評(píng)分,取材后測(cè)量結(jié)腸長(zhǎng)度,HE染色觀察結(jié)腸病理變化,ELISA法檢測(cè)結(jié)腸組織白介素-6(IL-6)、腫瘤壞死因子-α(TNF-α)水平,Western blot法檢測(cè)結(jié)腸組織中NF-κB、IκBα蛋白磷酸化水平,免疫熒光檢測(cè)結(jié)腸組織中NF-κB表達(dá)水平。結(jié)果:天樞穴組每日DAI評(píng)分顯著低于模型組(P0.01);結(jié)腸長(zhǎng)度針刺組顯著長(zhǎng)于模型組(P0.05);結(jié)腸組織HE染色觀察發(fā)現(xiàn)天樞穴組炎性損傷明顯輕于模型組;結(jié)腸組織中IL-6、TNF-α含量天樞穴組顯著低于模型組(P0.01),NF-κB、IκBα磷酸化水平亦明顯低于模型組(P0.01)。結(jié)論:針刺天樞穴可以明顯地改善DSS誘導(dǎo)IBD癥狀及結(jié)腸組織炎性反應(yīng)損傷程度,可能是通過(guò)抑制NF-κB信號(hào)通路而起到了治療作用,針刺天樞穴可以作為IBD一種有效的治療手段。
[Abstract]:Objective: to study the therapeutic effect of "Tianshu acupoint" on DSS induced colitis and its effect on NF-魏 B signal pathway. Methods: 30 C57BL / 6 mice were randomly divided into three groups: blank group, model group and Tianshu point group. The model group and Tianshu point group were given 3 DSS aqueous solution models respectively, and Tianshu acupoint group were treated with acupuncture for 20 minutes daily for 7 days. Daily DAI scores were recorded. The pathological changes of colon were observed by HE staining. The levels of interleukin-6 and TNF- 偽 in colon tissue were detected by Elisa and the phosphorylation of NF- 魏 B I 魏 B 偽 protein in colonic tissue was detected by Western blot. Results: the daily DAI score of Tianshu acupoint group was significantly lower than that of model group, the colonic length acupuncture group was significantly longer than that of model group (P 0.05), and the HE staining of colon tissue showed inflammatory lesion in Tianshu acupoint group. The injury was significantly lighter than that in the model group. The content of IL-6 TNF- 偽 in colon tissue in Tianshu group was significantly lower than that in model group. The phosphorylation level of I 魏 B 偽 in NF- 魏 B was also significantly lower than that in model group. Conclusion: acupuncture at Tianshu point can obviously improve the symptoms of IBD induced by DSS and the degree of inflammatory reaction injury in colon tissue. Acupuncture at Tianshu point may be an effective treatment for IBD by inhibiting NF- 魏 B signaling pathway.
