【摘要】：目的：通過研究苦柏顆粒洗劑在肛腸病術后肛門疼痛的療效,并對苦柏顆粒洗劑的鎮(zhèn)痛作用、作用原理和皮膚安全性進行研究,為在臨床中進行推廣提供科學依據。方法：采用隨機對照的方法,觀察苦柏顆粒洗劑治療疼痛的療效。將大鼠隨機分成5組,分別為空白組、1：5000高錳酸鉀溶液對照組、苦柏顆粒洗劑高、中、低劑量組。(1)大鼠足底熱板法,造模成功后,在最后一次涂抹藥物后的第30分鐘、60分鐘和90分鐘,通過統(tǒng)計大鼠在熱板儀上的舔足時間來比較各組藥物的鎮(zhèn)痛作用；(2)藥物注射扭體法,造模后分別在各組大鼠腹腔注射0.7%冰醋酸溶液,記錄下大鼠15分鐘內扭體反應出現(xiàn)的次數,來比較藥物的鎮(zhèn)痛效果；(3)ELISA法,對組大鼠造肛腸病術后疼痛模型,除模型對照組,其余各組切口分別給予相應藥物換藥,每日2次,治療3天后,目內眥取血,用ELISA法測定每組大鼠血清中IL-6和IL-10含量的改變；(4)皮膚安全性研究。結果：實驗結果表明,苦柏顆粒洗劑高劑量組、中劑量能提高大鼠足底的痛感覺閾值(P0.01或P0.05),也可以明顯抑制冰醋酸溶液所導致的大鼠扭體反應現(xiàn)象(P0.05)；苦柏顆粒高劑量組、中劑量(分別為24%和12%)能使肛腸病術后疼痛模型大鼠血清中的致痛物質白介素-6(IL-6)含量明顯降低、鎮(zhèn)痛物質白介素-10(IL-10)含量升高(P0.05或P0.01)；通過皮膚安全性實驗研究結果可見,苦柏顆粒洗劑經皮給藥,無紅腫瘙癢等反應。結論：苦柏顆粒洗劑對肛腸病術后肛門疼痛有確切的效果,療效強于對照組；苦柏顆粒洗劑局部給藥,有鎮(zhèn)痛作用,可以使血清中的IL-6降低、IL-10的含量升高；苦柏顆粒洗劑在皮膚局部給藥,無任何不良反應,安全性較高。
[Abstract]:Objective: to study the curative effect of Kubai granule lotion on anal pain after anorectal disease, and to study the analgesic effect, action principle and skin safety of Kubai granule lotion. Methods: the therapeutic effect of Kubai granule lotion on pain was observed by a randomized control method. Rats were randomly divided into five groups: blank group. 1: 5000 potassium permanganate solution control group, Kubai granule lotion high, medium, low dose group. 1) the rat plantar hot plate method, after the success of the model, 30 minutes after the last application of the drug. At 60 and 90 minutes, the analgesic effects of each group were compared by counting the time of licking foot on the hot plate. (2) to compare the analgesic effect of drug analgesia, 0.7% glacial acetic acid solution was injected intraperitoneally into each group of rats after drug injection and writhing method was used to record the times of writhing reaction in 15 minutes. The model of postoperative pain of anorectal disease was established in rats of group A by Elisa. Except for model control group, the other groups were given the corresponding drugs to change medicine twice a day, after 3 days of treatment, the blood was taken from the canthus of the eye. The changes of IL-6 and IL-10 in serum of each group were determined by ELISA method. Results: the results showed that the moderate dose of Kubai granule lotion could increase the threshold of pain sensation in the plantar of rats (P0.01 or P0.05). It also inhibited the writhing reaction induced by acetic acid solution in rats (P 0.05). In the high dose group, the middle dose (24% and 12, respectively) could significantly decrease the content of the pain-causing substance interleukin-6 in the serum of the model rats with postoperative pain of anorectal disease. The content of the analgesic substance interleukin-10 (IL-10) was higher than that of the control group (P 0.05 or P 0.01). The results of skin safety test showed that Kubai granule lotion had no reaction such as skin administration without redness and pruritus. Conclusion: Kubai granule lotion has definite effect on anal pain after anorectal disease, and the curative effect is better than that in control group. Local administration of Kubai granule lotion has analgesic effect, which can reduce the content of IL-10 in serum and decrease the content of IL-10. Kubai granule lotion is given locally in the skin without any adverse reaction and is safe.
