瑤藥銅鉆總黃酮對急性心肌缺血再灌注損傷保護作用的研究

【摘要】:目的:研究瑤藥銅鉆(Mappianthus iodoies Hand-Mazz)總黃酮對急性心肌缺血再灌注損傷的保護作用。方法:以72只成年健康雄性SD大鼠作為研究對象,隨機將其平均分為假手術組(Sham)、再灌注模型組(Model)、陽性藥物對照組(YXDZ)、瑤藥銅鉆總黃酮低劑量組(YYTZ-L 60 mg·kg-1)、瑤藥銅鉆總黃酮中劑量組(YYTZ-M 120 mg·kg-1)和瑤藥銅鉆總黃酮高劑量組(YYTZ-H 240 mg·kg-1)六個組。結扎大鼠左冠狀動脈前降支造成缺血30 min、解除結扎形成再灌注24 h的方法建立缺血再灌注模型。通過HE染色法對大鼠心肌組織病理形態(tài)學改變進行觀察;用TTC染色法檢測大鼠心肌梗死面積;ELISA試劑盒檢測大鼠血清中超氧化物歧化酶(SOD)、丙二醛(MDA)、腫瘤壞死因子-α(TNF-α)、白介素-6(IL-6)、肌酸激酶(CK)、乳酸脫氫酶(LDH)的濃度;免疫印跡檢測B細胞淋巴瘤/白血病-2(BCL-2)和BCL-2相關X蛋白(BAX)蛋白表達。結果:與Model相比,瑤藥銅鉆總黃酮劑量組對大鼠急性心肌缺血再灌注損傷的心肌組織病理形態(tài)學損害均有明顯改善,心肌梗死面積明顯較小(P0.05),血清中MDA、TNF-α、IL-6、CK、LDH含量均較低(P0.05),SOD含量較高(P0.05),BCL-2蛋白水平表達升高、BAX蛋白水平表達降低(P0.05);與Sham相比,Model心肌組織病理形態(tài)學損傷較明顯,心肌梗死面積明顯較大(P0.05),大鼠血清中CK、LDH、MDA、TNF-α、IL-6含量均明顯增高(P0.05),SOD含量較低(P0.05),大鼠BCL-2蛋白水平表達降低、BAX蛋白水平表達升高(P0.05)。結論:瑤藥銅鉆總黃酮對急性心肌缺血再灌注損傷有一定的保護作用,其機制可能與其抗炎、抗氧化及對抑制細胞凋亡作用有關。
[Abstract]:Objective: to study the iodoies Hand-Mazzs of Chinese herbal medicine Mappianthus iodoies Hand-Mazz. The protective effects of total flavonoids on acute myocardial ischemia-reperfusion injury were studied in 72 adult male SD rats. They were randomly divided into sham-operated group, reperfusion model group, and positive drug control group (YXDZ). YYTZ-L 60 mg 路kg-1). YYTZ-M (120 mg 路kg-1) and YYTZ-H (240 mg 路kg -1). The left anterior descending coronary artery was ligated for 30 min. The ischemia reperfusion model was established by removing ligation and forming reperfusion for 24 h. The pathological changes of myocardium in rats were observed by HE staining. Myocardial infarction area was measured by TTC staining. Serum superoxide dismutase (SOD), malondialdehyde (MDA), tumor necrosis factor- 偽 (TNF- 偽), interleukin-6 (IL-6) were detected by ELISA kit. The concentration of creatine kinase (CK) and lactate dehydrogenase (LDH); The expression of B-cell lymphoma / Leukemia BCL-2) and BCL-2 associated X protein was detected by Western blotting. Results: compared with Model. The total flavonoid dosage group of Yao medicine copper drill significantly improved the myocardial histopathologic damage of acute myocardial ischemia-reperfusion injury in rats, the myocardial infarction area was significantly smaller than that of P0.05, and MDA in serum was also significantly improved. The content of TNF- 偽, IL-6, CKL and LDH was higher than that of P0.05 and SOD, and the expression of BCL-2 protein was higher than that of P0.05 and BCL-2. The expression of BAX protein decreased in P0.05; Compared with Sham, myocardial histopathologic damage was more obvious, myocardial infarction area was significantly larger than P0.05, and serum CKD LDHDA-TNF- 偽 was found in rats. The content of IL-6 was significantly higher than that of P0.05, and the expression of BCL-2 protein was decreased in rats. Conclusion: the total flavonoids of Chinese herbal medicine have protective effect on acute myocardial ischemia-reperfusion injury, and its mechanism may be related to its anti-inflammatory mechanism.
