針灸與振動訓練對缺血缺氧性腦病大鼠細胞炎癥反應的研究

【摘要】:目的:觀察針灸與振動訓練對于缺血缺氧性腦病鼠炎癥反應的影響,初步探究針灸和振動訓練治療缺血缺氧性腦病的作用機制。方法:將成功建立缺血缺氧性腦病模型的60只SD大鼠,根據(jù)數(shù)字隨機表法分為4組,即模型對照組(不接受治療)、針刺組(僅接受針刺治療)、訓練組(僅接受振動訓練)以及聯(lián)合組(針刺聯(lián)合振動訓練)各15只,在建模前、建模后24h以及干預后2周時分別對大鼠神經功能進行評分,同時應用ELISA法檢測血清IL-1、IL-6表達水平,應用RT-PCR檢測腦組織中TNF-αmRNA、NF-κB mRNA表達水平,同時利用SPSS 19.0對實驗結果進行統(tǒng)計分析。結果:建模后,各組大鼠的神經行為評分、血清IL-1、IL-6、腦組織TNF-αmRNA、NF-κB mRNA顯著高于建模前(P0.05),但組間數(shù)據(jù)無明顯差異(P0.05);干預后,聯(lián)合組、針刺組、訓練組神經行為評分、血清IL-1、IL-6、腦組織TNF-αmRNA、NF-κB mRNA均明顯下降,且聯(lián)合組各指標下降最顯著,其次為針刺組,在干預2周時,各組上述指標差異顯著(P0.05)。結論:針灸聯(lián)合振動訓練能夠明顯減輕缺血缺氧性腦病細胞炎癥反應,其作用機制考慮與抑制IL-1、IL-6、TNF-α以及NF-κB等炎癥因子表達有關。
[Abstract]:Objective: To observe the effect of acupuncture and vibration training for hypoxic ischemic encephalopathy rat inflammatory response, preliminary study the mechanism of acupuncture and moxibustion and vibration training in the treatment of hypoxic ischemic encephalopathy. Methods: 60 SD rats of hypoxic ischemic encephalopathy model was established successfully, according to figures were randomly divided into 4 groups: model control group (no treatment), acupuncture group (received acupuncture treatment), training group (received vibration training) and combined group (acupuncture combined with vibration training) 15 rats each, before modeling, modeling after 24h and 2 weeks respectively after the intervention of the rat nerve function score, and serum IL-1 by ELISA method, the expression level of IL-6 and TNF- in brain tissue was detected using RT-PCR mRNA NF- kappa B alpha, mRNA expression level, while using SPSS 19 on experimental results were analyzed. Results: after modeling, the rats neurobehavioral score, serum IL-1, IL-6, brain group The fabric TNF- alpha mRNA NF- kappa B mRNA were significantly higher than that before modeling (P0.05), but no significant differences between groups of data (P0.05); intervention group, acupuncture group, training group, neurobehavioral score, serum IL-1, IL-6, TNF- in brain tissue mRNA NF- kappa B alpha, mRNA were significantly decreased, and the joint the group index decreased significantly, followed by acupuncture intervention, in 2 weeks, each of these indicators significantly difference (P0.05). Conclusion: acupuncture combined with vibration training can obviously reduce the hypoxic ischemic encephalopathy of inflammatory reaction, the mechanism of IL-6 and suppression of IL-1, NF-, and other inflammatory factor TNF- alpha kappa B expression.
