New Triterpene Prosaponin and Sapogenin with Unsaturated Lactone Skeleton From Gynostemma Pentaphfllum and the Antitumor Activities Assay

【摘要】：正Object:Studying the anti-tumor activities of the prosaponin and sapogenin of ginsenoside from Gynostemma pentaphfllum in order to find more active lead compound and investigate their structure-activity relationship.Method:Selective acid hydrolysis,and repeated silica gel column chromatography and preparation lamellar were used to obtain the prosaponin and sapogenins.And and the 1D(~1H NMR,~(13)C NMR),2D spectroscopic analysis were used for the structural identification.In vitro cytotoxicity bioassay were employed using MTT method.Result: A novel prosaponin(3β,23S)-3-O-β-D-glucopyranosyl-23-hydroxyldammar-20,24-dien-21-oic acid-21,23-lactone(1),together with 3 new sapogenins(2,3,4)with unsaturated lactone skeleton and 5 known sapogenins 20(S)-panaxadiol(5),20(R)-protopanaxadiol(6),24 (R)-ocotillol(7),20(R)-dammarane-3β,12β,20,25-tetrol(8),20(R)-dammarane-3β,6α,12β,20, 25-pentol(9)have been isolated from the hydrolyzed saponins of Gynostemma pentaphfllum.Their structures were identified by spectroscopic methods including NMR,IR and MS,and their cytotoxicity was evaluated against four kinds of tumor cell lines.Conclusion:The novel prosaponin(1)showed strong antitumor activity against HL-60 cell lines and mild activities against other five kinds of human tumor cell lines.
