【摘要】：目的：通過比較尋常型銀屑病血熱證患者外周血中Th17、Treg細(xì)胞的特異性轉(zhuǎn)錄因子RORγt、Foxp3mRNA的表達(dá)與健康對(duì)照組的差異,分析其與血熱證證型的相關(guān)性,以期為尋常型銀屑病血熱證的微觀辨證做一些補(bǔ)充性研究。方法：采用實(shí)時(shí)熒光定量逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(real-time RT-PCR),對(duì)尋常型銀屑病血熱證患者外周血中Th17、Treg細(xì)胞的特異性轉(zhuǎn)錄因子RORγt、 Foxp3mRNA的表達(dá)水平進(jìn)行量化分析。結(jié)果：1、尋常型銀屑病血熱證組30例外周血中Th17、Treg細(xì)胞的特異性轉(zhuǎn)錄因子RORγt, Foxp3mRNA相對(duì)表達(dá)量分別為1.99±0.81,1.13±0.56,分別與正常對(duì)照組相比,差異有統(tǒng)計(jì)學(xué)意義(t值分別為2.50,2.57,P均0.05),說明患病組外周血中RORγtmRNA的表達(dá)量明顯高于對(duì)照組,Foxp3mRNA的表達(dá)量則明顯低于對(duì)照組；2、尋常型銀屑病血熱證組30例外周血中RORγt/Foxp3表達(dá)量的比值為1.04±0.77,與正常對(duì)照組相比,差異無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論：1、尋常型銀屑病血熱證患者外周血中Th17細(xì)胞轉(zhuǎn)錄因子RORγtmRNA與健康人相比較呈高表達(dá),可以佐證Th17細(xì)胞參與了尋常型銀屑病血熱證證候的產(chǎn)生；尋常型銀屑病血熱證患者外周血中Treg細(xì)胞轉(zhuǎn)錄因子Foxp3mRNA與健康人相比呈低表達(dá),可以佐證Treg細(xì)胞參與了尋常型銀屑病血熱證證候的產(chǎn)生。但轉(zhuǎn)錄因子RORγ、Foxp3僅僅是Th17、Treg細(xì)胞參與發(fā)病的一小部分,從多層次、多方位研究Th17、Treg細(xì)胞與尋常型銀屑病血熱證證候相關(guān)性需要我們進(jìn)一步去探索；2、尋常型銀屑病血熱證患者轉(zhuǎn)錄因子RORγt/Foxp3表達(dá)與健康人比較沒有統(tǒng)計(jì)學(xué)意義,但近幾年很多資料證實(shí)Th17/Treg細(xì)胞失衡是尋常型銀屑病發(fā)病的關(guān)鍵,RORγt是Th17細(xì)胞轉(zhuǎn)錄因子,Foxp3是Treg細(xì)胞轉(zhuǎn)錄因子,RORγt/Foxp3表達(dá)比值不能完全反映Th17/Treg細(xì)胞失衡的情況。
[Abstract]:Objective: to compare the expression of ROR 緯 ttcr3 mRNA in peripheral blood of patients with blood fever of psoriasis vulgaris. Analysis of its correlation with the type of blood heat syndrome. Objective: to do some supplementary research on micro differentiation of blood fever syndrome of psoriasis vulgaris. Methods: Real-time RT PCR was used to detect Psoriasis vulgaris. The expression of ROR 緯, Foxp3mRNA in peripheral blood of patients with psoriasis vulgaris was quantitatively analyzed. The relative expression of ROR 緯 t and Foxp3mRNA in peripheral blood of 30 patients with blood fever of psoriasis vulgaris was 1.99 鹵0.81, respectively. 1.13 鹵0.56, compared with the normal control group, the difference was statistically significant (2.50 鹵2.57g, P < 0.05). The results showed that the expression of ROR 緯 tmRNA in the peripheral blood of the diseased group was significantly higher than that of the control group, and the expression of Foxp3 mRNA was significantly lower than that of the control group. 2, the ratio of ROR 緯 t / Foxp3 expression in peripheral blood of 30 patients with psoriasis vulgaris was 1.04 鹵0.77, compared with that of normal controls. Conclusion: Th17 transcription factor ROR 緯 tmRNA in peripheral blood of patients with blood fever of psoriasis vulgaris is higher than that of healthy persons. It can be proved that Th17 cells are involved in the production of blood fever syndrome of psoriasis vulgaris. The expression of Treg transcription factor Foxp3mRNA in peripheral blood of patients with blood fever of psoriasis vulgaris was lower than that of healthy people. It can be concluded that Treg cells are involved in the production of blood fever syndrome of psoriasis vulgaris, but the transcription factor ROR 緯 Foxp3 is only a small part of Th17T Treg cells involved in the pathogenesis of psoriasis vulgaris. The relationship between Th17 Treg cells and blood fever syndrome of psoriasis vulgaris needs to be further explored. 2. The expression of transcription factor ROR 緯 t / Foxp3 in patients with blood fever of psoriasis vulgaris was not significantly different from that in healthy persons. However, in recent years, many data have confirmed that Th17/Treg cell imbalance is the key to the pathogenesis of psoriasis vulgaris. ROR 緯 t is a transcription factor of Th17 cells and Foxp3 is a transcription factor of Treg cells. The ratio of ROR 緯 t / Foxp3 expression can not completely reflect the imbalance of Th17/Treg cells.
