蒙藥復(fù)方述達(dá)格-4的大鼠血清化學(xué)成分分析

【摘要】:目的建立蒙藥復(fù)方述達(dá)格-4體內(nèi)外特征圖譜,歸屬并鑒定其體外化學(xué)成分及體內(nèi)吸收入血成分。方法取Wistar大鼠,灌胃蒙藥復(fù)方述達(dá)格-4及其各單味藥有效組分,分離制備含藥血清,HPLC法測(cè)定,建立蒙藥復(fù)方述達(dá)格-4體外及含藥血清HPLC特征圖譜,分析蒙藥復(fù)方述達(dá)格-4各單味藥材體外及其大鼠含藥血清特征圖譜,比對(duì)、歸屬并鑒定蒙藥復(fù)方述達(dá)格-4體外化學(xué)成分及口服蒙藥復(fù)方述達(dá)格-4后的大鼠吸收入血成分。結(jié)果分析蒙藥復(fù)方述達(dá)格-4體外HPLC特征圖譜,共發(fā)現(xiàn)了64個(gè)峰。蒙藥復(fù)方述達(dá)格-4灌胃給藥后出現(xiàn)了23個(gè)入血成分,17個(gè)為體外成分直接入血,另外6個(gè)推測(cè)為新生代謝產(chǎn)物。蒙藥復(fù)方述達(dá)格-4灌胃給藥后出現(xiàn)的23個(gè)入血成分全部來(lái)源于各單味藥材及全方,其中1,2,3,9,23號(hào)峰來(lái)源于高良姜;14號(hào)峰來(lái)源于木香;13號(hào)峰來(lái)源于石菖蒲;15,20,21,22號(hào)峰來(lái)源復(fù)雜,均來(lái)源于高良姜、木香、石菖蒲;6,10,17,18,19號(hào)峰來(lái)源于高良姜、石菖蒲;7,16號(hào)峰來(lái)源于高良姜、木香;11號(hào)峰來(lái)源木香、石菖蒲;4,5,8,12號(hào)峰來(lái)源于全方,并從中鑒定出7個(gè)蒙藥復(fù)方述達(dá)格-4成分。結(jié)論血中成分及代謝產(chǎn)物可能為蒙藥復(fù)方述達(dá)格-4體內(nèi)直接作用的有效成分,主要來(lái)源于蒙藥復(fù)方述達(dá)格-4及其各單味藥。
[Abstract]:Objective to establish the dagonet Mongolian compound -4 in vitro and in vivo fingerprint, belonging and identification of the chemical constituents in vitro and in vivo absorption into the blood. Methods Wistar rats, intragastric administration of the DAG Mongolian compound -4 and each single herb effective components, separation and preparation of medicated serum HPLC determination, the establishment of Mongolian medicine compound DAG -4 in vitro and serum HPLC fingerprint analysis, the DAG -4 Mongolian compound each single medicine containing serum and in vitro characteristics of rat atlas, alignment, belonging and identification of Mongolian medicine compound -4 in vitro the DAG chemical composition and oral Mongolian medicine compound the DAG after -4 rats absorbed in blood. The results of the analysis of the Mongolian medicine DAG -4 in vitro HPLC fingerprint, found a total of 64 peaks. The Mongolian compound DAG -4 intragastric administration after 23 into the blood components, the 17 components for in vitro directly into the blood, the other 6 presumed new Mongolian compound metabolites. The DAG -4 after oral administration of the 23 components into the blood from all of each herb and the whole party, the 1,2,3,9,23 peak from Gaoliangjiang; the No. 14 peak to peak 13 from costusroot; Shichangpu; 15,20,21,22 peaks are derived from complex sources, galangal, woody, Shichangpu 6,10,17,18,19; peak from galangal, Shichangpu; 7,16 peak from Rhizoma radix; the No. 11 peak woody, Shichangpu; 4,5,8,12 peak comes from the whole party, and 7 of the DAG Mongolian compound -4 components from identified constituents and metabolites. Conclusion in blood may be the effective ingredient of Mongolian medicine the DAG -4 in vivo direct action, mainly from the Mongolian compound -4 and DAG of each drug.
