【摘要】:目的對尋常型銀屑病(血熱證、血瘀證、血虛證)患者與正常人差異蛋白進行分析,探求三證之間差異性表達蛋白,從蛋白質組學角度揭示三證之間的差異。方法取尋常型銀屑病血瘀證、血虛證、血熱證患者的血清,應用蛋白提取、定量、雙向電泳技術、質譜鑒定和數(shù)據(jù)庫檢索,尋找出三證之間的差異蛋白和共有蛋白,并將這些蛋白進行組間比較。結果發(fā)現(xiàn)了15種顯著性差異蛋白(P0.05),其中補體C3、凝聚素、血清轉鐵蛋白、間-α-胰蛋白酶抑制劑H4重鏈(片段)、載脂蛋白E、補體C4-B、甘露糖結合蛋白C及富亮氨酸-α-2-糖蛋白在血虛型銀屑病患者血清表達水平高于血熱型和血瘀型(P0.05)。纖維蛋白原β鏈、血液接合素、纖維蛋白原γ鏈、HPX蛋白、α-抗胰蛋白酶、甲狀腺素轉運蛋白及載脂蛋白Ⅳ在血熱型銀屑病患者血清中的表達水平高于血瘀型患者(P0.05)。共同蛋白有5種(P0.05),分別為血液結合素、纖維蛋白原γ鏈、HPX蛋白、α-抗胰蛋白酶、載脂蛋白A-Ⅳ。結論尋常型銀屑病(血熱證、血瘀證、血虛證)血清中存在表達差異性蛋白質,可能與尋常型銀屑病的發(fā)病機制和銀屑病不同證型的物質基礎有關。
[Abstract]:Objective to analyze the differential protein between patients with psoriasis vulgaris (blood heat syndrome, blood stasis syndrome, blood deficiency syndrome) and normal people, and to explore the differential expression of protein between the three syndromes. Methods Blood stasis syndrome, blood deficiency syndrome and blood heat syndrome of psoriasis vulgaris were collected from the serum of patients with blood stasis syndrome, blood deficiency syndrome and blood heat syndrome, and protein extraction, quantitative analysis and two dimensional electrophoresis were used to detect the differences between the three syndromes from the perspective of proteomics. The differential proteins and common proteins among the three syndromes were identified by mass spectrometry and database retrieval. The results showed that there were 15 significant differential proteins (P0.05). Among them, complement C3, agglutinin, serum transferrin, m-偽 -trypsin inhibitor H4 heavy chain (fragment, apolipoprotein E, complement C4-B). The expression level of mannose binding protein C and leucine-rich 偽 -2-glycoprotein in serum of psoriasis patients with blood deficiency was higher than that of P0.05, fibrinogen 尾 chain and blood conjugate in psoriatic patients with blood deficiency.
