氣虛痰濁證高血壓大鼠動(dòng)物模型研究

【摘要】:目的擬構(gòu)建氣虛痰濁證高血壓大鼠模型,為中醫(yī)藥研究提供載體。方法將50只12周齡自發(fā)性高血壓大鼠(spontaneously hypertensive rats,SHRs),隨機(jī)分為模型組(40只)及對照組(10只),同時(shí)設(shè)置10只12周齡正常血壓大鼠(wistar-Kyoto,WKY)為對照(WKY組)。模型組隔日喂食高脂飼料,每周6次力竭游泳;對照組及WKY組始終以普通大鼠飼料喂養(yǎng)。對各組大鼠表征體系進(jìn)行動(dòng)態(tài)觀察,12周后進(jìn)行相關(guān)指標(biāo)檢測。結(jié)果模型組大鼠隨著造模時(shí)間延長,分別出現(xiàn)生物學(xué)表征評分升高、游泳時(shí)間縮短、體質(zhì)量增加、曠場分析中央格停留時(shí)間延長、水平運(yùn)動(dòng)距離縮短、血壓升高等;血清指標(biāo)檢測中,模型組甘油三酯(TC)、膽固醇(TG)、血糖(GLU)均高于其他2組(P0.01);模型組低密度脂蛋白(LDL-C)較WKY組升高(P0.05);模型組雙側(cè)頸動(dòng)脈血流量比對照組、WKY組降低,差異有統(tǒng)計(jì)學(xué)意義(P0.01)。結(jié)論以SHRs為基礎(chǔ),持續(xù)力竭游泳、隔日禁食、高脂飼料喂養(yǎng)建立氣虛痰濁證高血壓大鼠模型,相關(guān)表征及血清指標(biāo)改變符合氣虛痰濁證候特點(diǎn)。
[Abstract]:Objective to construct the model of hypertension rats with Qi deficiency and phlegm turbidity syndrome, and to provide the carrier for the research of traditional Chinese medicine. Methods a total of 50 12 week old spontaneously hypertensive rats (spontaneously hypertensive rats, SHRs) were randomly divided into model group (40 rats) and control group (10 rats), and 10 12 week old normotensive rats (wistar-Kyoto, WKY) were served as control group (WKY group). The model group fed high fat diet every other day, swimming 6 times a week, and the control group and the WKY group were always fed with the common rat feed. The dynamic observation was carried out in each group of rats, and the related indexes were detected after 12 weeks. The rats in the model group with the modeling time, respectively. The biological characterization score was increased significantly, swimming time, increased body weight, open field test, the central grille time, horizontal movement distance shortened, high blood pressure; serum marker, glycerol three in the model group (TC) and cholesterol ester (TG), blood glucose (GLU) were higher than the other 2 groups (P0.01); model group, low density lipoprotein (LDL-C) was higher than that in WKY group (P0.05 group); carotid artery blood flow model than the control group, WKY group decreased, the difference was statistically significant (P0.01). Conclusion based on SHRs, a rat model of qi deficiency, phlegm turbid syndrome and hypertension is established based on continuous exhaustive swimming, fasting and high-fat diet. The related characteristics and serum indexes are in accordance with the characteristics of qi deficiency, phlegm turbidity syndrome.
