枇杷葉不同組分提取物在多種疾病中的作用及機(jī)制研究(英文)

目的提取枇杷葉中的多糖、黃酮和三萜酸,并分別研究其在抗炎、抗氧化和抗腫瘤中的藥理活性。方法通過(guò)水提醇沉法提取多糖,醇沉上清液用于黃酮的富集,水提的殘?jiān)?jīng)90%乙醇回流提取三萜酸。通過(guò)Western blot檢測(cè)枇杷葉多糖對(duì)TGF-β1干預(yù)的小鼠系膜細(xì)胞中PAI-1表達(dá)的影響,觀察枇杷葉中的黃酮成分對(duì)ABTS自由基的清除作用評(píng)價(jià)其抗氧化活性;MTT法檢測(cè)枇杷葉中三萜酸對(duì)骨髓瘤細(xì)胞ARP1增殖的影響,并通過(guò)ARP1細(xì)胞小鼠異種移植模型觀察三萜酸對(duì)皮下腫瘤生長(zhǎng)的影響。結(jié)果枇杷葉中多糖、黃酮、三萜酸的含量分別為19.53、48.70、7.86 g/kg。枇杷葉多糖可以抑制PAI-1表達(dá),黃酮濃度在75~150μg/mL對(duì)ABTS自由基具有良好的清除作用,三萜酸可以抑制骨髓瘤細(xì)胞ARP1的增殖,IC50為11.57μg/mL。此外,三萜酸對(duì)異種移植小鼠皮下腫瘤的生長(zhǎng)具有一定的抑制作用。結(jié)論枇杷葉中多糖可能對(duì)腎炎具有一定的治療作用,黃酮和三萜酸具有一定的抗氧化和抗腫瘤的作用。為枇杷葉的進(jìn)一步開(kāi)發(fā)利用提供有利的實(shí)驗(yàn)基礎(chǔ)。
