白藜蘆醇抗小鼠急性T淋巴細胞白血病作用及Notch1信號通路機制的研究

目的:觀察白藜蘆醇對急性T淋巴細胞白血病小鼠的作用及Notch1、Hes-1信號分子的表達水平,并探討其作用機制。方法:1.完全隨機法將50只C57BL/6雌性健康小鼠分為5組:正常對照組;模型對照組;白藜蘆醇低濃度處理組(25mg/kg/d);白藜蘆醇中濃度處理組(50mg/kg/d);白藜蘆醇高濃度處理組(100mg/kg/d)。尾靜脈注射1×106個GFP+白血病細胞,制備T-ALL白血病小鼠模型。2.應用瑞氏-吉姆薩染色觀察各組外周血白血病細胞浸潤情況。3.流式細胞術檢測各實驗組外周血、脾細胞懸液中白血病細胞的百分比。4.HE染色法觀察各實驗組肝臟、脾臟、腎臟、骨髓組織中白血病細胞的浸潤情況。5.應用實時熒光定量PCR(RT-PCR)檢測各組脾臟組織中Notch1、Hes-1信號分子mRNA的表達量。6.應用免疫蛋白印跡法(Western Blot)檢測各組脾臟組織中Notch1、Hes-1蛋白表達水平。結(jié)果:1.模型對照組、白藜蘆醇低、中、高濃度處理組的外周血涂片均可觀察到原始淋巴細胞,其細胞核呈圓形或略呈卵圓形,大而深染,染色質(zhì)致...
