ATM介導(dǎo)白藜蘆醇改善高脂誘導(dǎo)胰島素抵抗及骨骼肌糖脂代謝紊亂的研究

流行病學(xué)資料顯示,肥胖及2型糖尿。╰ype 2 diabetes mellitus,T2DM)等代謝性疾病已成為全球公共健康問題,這些代謝性疾病重要的病理基礎(chǔ)之一是胰島素抵抗(insulin resistance,IR),而骨骼肌IR是全身IR的重要病因之一。近年我國飲食習(xí)慣受到西方文化的影響,高脂高熱量食物消耗量明顯增加。T2DM及肥胖與不良的飲食和作息習(xí)慣,尤其是脂肪的大量攝入有關(guān),但其具體機(jī)制尚不十分清楚。因此,高脂飲食導(dǎo)致的IR或脂質(zhì)沉積是較好的研究其機(jī)制和尋求治療方法的模型。我們課題組的前期研究發(fā)現(xiàn)高脂能夠誘導(dǎo)大鼠及小鼠的IR模型,并伴有骨骼肌脂質(zhì)沉積。因此,本研究選取了高脂誘導(dǎo)的IR模型作為實(shí)驗(yàn)?zāi)P。白藜蘆醇(resveratrol,Rsv)是一種多酚類植物物質(zhì),被稱為天然的“熱量限制類似物”。近些年Rsv因其抗炎、抗氧化、改善胰島素敏感性及抗腫瘤等多種功效日益受到人們的關(guān)注,但具體機(jī)制存在爭(zhēng)議,仍有待深入研究。共濟(jì)失調(diào)毛細(xì)血管擴(kuò)張癥突變(ataxia-telangiectasia-mutated,ATM)基因是共濟(jì)失調(diào)毛細(xì)血管擴(kuò)張癥(ataxia-telangiecta...
