模擬失重30 d后再超重對猴牙齦組織趨化因子CCL20及其受體CCR6表達的影響

【摘要】:目的:探討模擬失重30 d后再超重對猴牙齦組織趨化因子CCL20及其受體CCR6表達的影響。方法:23只雄性獼猴隨機分為4組,即對照組(A)、失重組(B)、超重組(C)、模擬失重30 d后再超重組(D)。其中,D組根據(jù)載荷量大小又分為4個亞組,即+11Gx/270 s組(D1)、+13Gx/230 s組(D2)、+15 Gx/200 s組(D3)和+13 Gx/230 s恢復9 d組(D4)。采用組織學方法觀察牙齦的形態(tài)學變化,通過免疫組織化學、Q-PCR方法檢測牙齦組織中CCL20、CCR6的表達情況。采用SPSS17.0軟件包對數(shù)據(jù)進行統(tǒng)計學分析。結果:各實驗組中牙齦結構均未見明顯損傷,但實驗組中的淋巴細胞及中性粒細胞浸潤較對照組多。對照組牙齦組織幾乎不表達CCL20,但弱表達CCR6;實驗組CCL20陽性表達主要位于牙齦上皮組織的基底層,CCR6陽性表達主要位于牙齦上皮棘層和基底層。除失重組的CCL20外,各實驗組牙齦組織中的CCL20和CCR6在蛋白水平、mRNA水平上的表達均顯著高于對照組(P0.05)。結論:模擬失重30 d后再超重環(huán)境中,猴牙齦組織結構未見明顯改變,但牙齦組織趨化因子CCL20及其受體CCR6表達明顯增強。
[Abstract]:Aim: to investigate the effect of overweight on the expression of chemokine CCL20 and its receptor CCR6 in gum tissue of monkeys after 30 days of simulated weightlessness. Methods: 23 male rhesus monkeys were randomly divided into 4 groups: control group (A), lost recombinant (B), superrecombinant (C), simulated weightlessness for 30 days, and then superrecombinant (D). Among them, group D was divided into four subgroups according to the load size, namely, 11Gx/270 s group (D1), 13Gx/230 s group (D2), 15 Gx/200 s group (D 3) and 13Gx/230 s recovery 9 d group (D4). The morphological changes of gums were observed by histology, and the expression of CCL20,CCR6 in gum tissues was detected by immunohistochemistry and Q-PCR. SPSS17.0 software package was used to analyze the data statistically. Results: there was no obvious damage to gum structure in each experimental group, but the infiltration of lymphocytes and neutrophils in the experimental group was more than that in the control group. In the control group, there was almost no expression of CCL20, in gum tissue, but the positive expression of CCL20 in the experimental group with weak expression of CCR6; was mainly located in the basal layer of the gum epithelial tissue, and the positive expression of CCR6 was mainly located in the spinous layer and basal layer of the gum epithelial tissue. Except for the lost CCL20, the expression of CCL20 and CCR6 in the gum tissue of each experimental group was significantly higher than that of the control group at the protein level and mRNA level (P 0.05). Conclusion: in the overweight environment after 30 days of simulated weightlessness, the gum tissue structure of monkeys did not change significantly, but the expression of chemokine CCL20 and its receptor CCR6 in gum tissue was significantly increased.
