文章摘要:研究人體腸道細(xì)菌對(duì)阿魏酸的代謝。采集健康志愿者新鮮糞便,稱重稀釋后厭氧培養(yǎng),通過反復(fù)涂布分離得到69種人體腸道細(xì)菌。采用超高效液相四級(jí)桿飛行時(shí)間串聯(lián)質(zhì)譜技術(shù)(UPLC-Q-TOF/MS)與MetabolynxTM軟件分析阿魏酸經(jīng)人體糞便混合菌及分離的單菌作用后的代謝產(chǎn)物,共鑒定出了3種代謝產(chǎn)物,分別是氫化產(chǎn)物、去甲基化產(chǎn)物及去甲氧基化產(chǎn)物;此外,篩選出了對(duì)阿魏酸有特殊代謝活性的2種單菌,其中一種為已鑒定了的Bacillus sp.46;而另一種未知的細(xì)菌則用16S核糖體RNA(16S rRNA)技術(shù)進(jìn)行了鑒定,結(jié)果表明,此細(xì)菌為Enterococcus sp.45。本研究提示腸道細(xì)菌對(duì)阿魏酸有代謝作用且由不同細(xì)菌完成。
Abstr:The investigation on the metabolism of ferulic acid by incubating ferulic acid with different isolated bacteria from human feces in vitro was carried out. Fresh human fecal sample was obtained from a healthy volunteer, diluted serially in sterile water and sixty-nine different bacterial colonies were picked out ultimately. Ultra performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q- TOF/MS) with automated data analysis software ( MetaboLynxTM ) was applied to fast analysis of [erulic acid metabolites. The results indicated that hydrogenation, demethylation and demethoxylation were the major metabolic pathways of ferulic acid by human intestinal bacteria in vitro. Two strain bacteria were found to metabolize ferulic acid and one of them had been identified as Bacillus sp. 46. The 16S rRNA gene of the other isolated bacterium sp. 45 was base pair sequences and its nucleotide sequence has registered in GenBank under accession no. KC41732. Finally, this strain was identified as Enterococcus sp. 45 by phylogenetic affiliation. This study suggested that intestinal bacteria had the metabolic effects of ferulic acid and the biotransformation was completed by different bacteria.
