神經(jīng)節(jié)苷脂對急性低氧大鼠腦皮層神經(jīng)元的保護作用研究

【摘要】:目的觀察外源性神經(jīng)節(jié)苷脂對急性減壓低氧大鼠腦神經(jīng)元的保護作用 ,為臨床應(yīng)用提供依據(jù)。方法 1 5只Wistar大鼠隨機分為 :低氧組A、低氧組B與常氧組。低氧組A置于海拔 70 0 0m ,持續(xù) 5h ,低氧組B在低氧 3d前腹腔注射神經(jīng)節(jié)苷脂 ( 1 0 0mg/kg/d) ,其余與低氧組A相同。電鏡觀察各組腦皮層神經(jīng)元的超微結(jié)構(gòu)改變。結(jié)果低氧組B動物的腦神經(jīng)元胞體、細(xì)胞骨架和突觸結(jié)構(gòu)損傷的程度明顯輕于低氧組A。結(jié)論神經(jīng)節(jié)苷脂對低氧性腦損傷有一定的保護作用 ,這可能與其減少低氧時胞內(nèi)Ca2 +蓄積和穩(wěn)定CaM水平與CaMKinaseⅡ活性變化有關(guān)。
[Abstract]:Objective to observe the protective effect of exogenous ganglioside on brain neurons in rats with acute decompression and hypoxia, and to provide evidence for clinical application. Methods Fifteen Wistar rats were randomly divided into hypoxia group, hypoxia group B and normoxic group. Hypoxia group A was placed at 7 000 m above sea level for 5 hours. Rats in hypoxia group B were injected with ganglioside (10 0mg/kg/d) 3 days before hypoxia, and the rest were the same as those in hypoxia group A. Ultrastructural changes of cortical neurons in each group were observed by electron microscope. Results the degree of damage to the neuronal cell body, cytoskeleton and synaptic structure in hypoxic group B was significantly less than that in hypoxic group A. Conclusion ganglioside has a protective effect on hypoxic brain injury, which may be related to the decrease of intracellular Ca2 accumulation and stable CaM level and the change of CaMKinase 鈪,
