冷暴露大鼠血漿腎上腺素及血管內(nèi)皮細胞NOS活性的變化

【摘要】:目的觀察冷暴露大鼠血漿腎上腺素(Adr)水平及血管內(nèi)皮細胞NOS活性的變化,為探尋防治冷損傷的措施提供理論依據(jù)。方法使用L-精氨酸(L-Arg)調(diào)節(jié)主動脈內(nèi)皮細胞NOS活性。大鼠隨機分為5組,即對照組(室溫)、冷暴露Ⅰ組(-15℃,1 h)、冷暴露Ⅱ組(-15℃,1.5 h)、L-Arg組(室溫,L-Arg 2 g.kg-1灌服)、冷暴露+L-Arg組(-15℃,1 h;L-Arg 2 g.kg-1灌服)。冷暴露后6 h再取血樣。以酶聯(lián)免疫法測定血漿腎上腺素水平;以硝酸酶還原法測定血管內(nèi)皮細胞NOS活性;以紫外分光光度計檢測血清和血管內(nèi)皮細胞培養(yǎng)液中的LDH活性;反轉(zhuǎn)錄PCR方法檢測血管內(nèi)皮細胞NOS mRNA表達水平。結(jié)果對照組、冷暴露Ⅰ組與Ⅱ組血漿中Adr分別為(10.81±1.85)、(31.37±4.48)、(39.35±5.59)nmol.L-1,冷暴露大鼠血漿中Adr水平明顯地高于對照組(P0.05);上述3組血管內(nèi)皮細胞NOS活性分別為(16.13±3.68)、(9.19±1.87)、(5.94±1.05)μmol.L-1;冷暴露大鼠血管內(nèi)皮細胞NOS活性明顯低于對照組(P0.05),使用L-Arg可使冷暴露大鼠血管內(nèi)皮細胞NOS活性上調(diào)到(12.87±1.60)μmol.L-1(P0.01)。同時,血漿中LDH水平也明顯降低(P0.01)。結(jié)論冷暴露血漿高濃度腎上腺素可抑制主動脈內(nèi)皮細胞NOS活性;L-Arg可上調(diào)血管內(nèi)皮細胞NOS活性,對減輕機體冷損傷程度有一定作用。
