【摘要】：目的分析 5個(gè)與離心機(jī)訓(xùn)練有關(guān)的基因在大鼠各組織的表達(dá)水平及其與訓(xùn)練時(shí)間的關(guān)系。方法分別提取離心機(jī)訓(xùn)練不同時(shí)間組大鼠心、腦、腎、肺和腸 5種組織的mRNA并進(jìn)行量化 ,采用狹線雜交方法 ,用Dig 1 1 dUTP標(biāo)記 5個(gè)用抑制性消減雜交篩選離心機(jī)訓(xùn)練大鼠獲得的基因片段作探針 ,雜交結(jié)果通過光密度掃描半定量后 ,進(jìn)行統(tǒng)計(jì)學(xué)處理。結(jié)果未知基因CH1 5 7的表達(dá)水平在訓(xùn)練 6d時(shí)大鼠心臟和腦組織明顯下降 (P 0 .0 5 ) ,訓(xùn)練 1 2d時(shí)有所回升 ;未知基因CH2 4 4的表達(dá)水平在大鼠心、腦、腸組織在訓(xùn)練 6d、1 2d時(shí)均下調(diào) ,但其表達(dá)量個(gè)體差異表現(xiàn)突出 ,不呈現(xiàn)明顯的統(tǒng)計(jì)學(xué)意義 ;神經(jīng)元一氧化氮合酶抑制劑 (PIN)基因 ,隨著離心機(jī)訓(xùn)練時(shí)間的延長 ,在大鼠心臟和腎臟組織中的表達(dá)水平明顯上調(diào)(P 0 .0 5 ) ;細(xì)胞色素b (Cytb)基因在大鼠心臟、腦組織中表達(dá)水平隨著訓(xùn)練時(shí)間增長而明顯上調(diào) (P0 .0 1 ) ,在腎、肺和腸組織中的表達(dá)在訓(xùn)練 6d時(shí)明顯升高 (P 0 .0 5 ) ,訓(xùn)練 1 2d時(shí)有所恢復(fù) ;延長因子 1 α(EF1 α)基因在大鼠腦組織中表達(dá)水平隨著訓(xùn)練時(shí)間增長而明顯上調(diào) ,在腸組織中的表達(dá)在訓(xùn)練 6d時(shí)明顯升高 (P 0 .0 5 ) ,訓(xùn)練 1 2d時(shí)恢復(fù)到訓(xùn)練前水平。結(jié)論 5個(gè)基因在離心機(jī)訓(xùn)練大鼠不同組織中的表達(dá)水平存在差異 ,
[Abstract]:Objective to analyze the expression level of five genes related to centrifuge training in rat tissues and their relationship with training time. Methods the mRNA of heart, brain, kidney, lung and intestine of rats trained with centrifuge for different time groups were extracted and quantified. Dig 11 dUTP was used to label 5 gene fragments obtained from centrifuge training by suppression subtractive hybridization as probes. The hybridization results were processed by statistical analysis after semi-quantitative optical density scanning. Results the expression level of unknown gene CH1 57 was significantly decreased in the heart and brain of rats at 6 days of training (P 0. 05) and increased at 12 days, and the expression level of unknown gene CH2 44 was found in the heart and brain of rats. The intestinal tissue was down-regulated at 6 days and 12 days after training, but the expression level of intestinal tissue was significantly different from that of the control group, and there was no significant difference between them. The neuronal nitric oxide synthase inhibitor (PIN) gene increased with the training time of centrifuge. The expression of cytochrome b (Cytb) gene in rat heart and brain tissues increased significantly with the training time (P0. 01), and the expression level of cytochrome b (Cytb) gene in the rat heart and brain tissues increased significantly (P0. 01), and the expression of cytochrome b (Cytb) gene in the rat heart and brain tissue increased significantly (P0. 01) with the increase of training time (P0. 01). The expression of EF1 偽 in lung and intestine increased significantly at 6 days of training (P 0. 05) and recovered after 12 days of training, and the expression of EF1 偽 gene increased with the increase of training time. The expression level in intestinal tissue increased significantly at 6 days training (P 0. 05), and recovered to pre training level at 12 days training. Conclusion there are differences in the expression levels of five genes in different tissues of centrifuge training rats.
