【摘要】：為探討模擬失重對心肌成纖維細(xì)胞Ⅰ型膠原代謝的影響 ,本研究采用回轉(zhuǎn)器模擬失重效應(yīng) ,通過免疫細(xì)胞化學(xué)和反轉(zhuǎn)錄聚合酶鏈?zhǔn)椒磻?yīng) (RT PCR)研究了回轉(zhuǎn)模擬失重對原代培養(yǎng)的新生大鼠心肌成纖維細(xì)胞Ⅰ型膠原蛋白及mRNA表達(dá) ,以及膠原降解抑制物———金屬蛋白酶組織抑制因子 (Tissueinhibitorofmetallopro teinase ,TIMP)mRNA表達(dá)的影響。免疫細(xì)胞化學(xué)染色顯示回轉(zhuǎn)組Ⅰ型膠原蛋白沉積增加 ;RT PCR分析顯示Ⅰ型膠原α1鏈 (TypeⅠcollagenα1chain ,ColⅠA1)mRNA表達(dá)沒有明顯變化 ,TIMP 1、TIMP 2及TIMP 3的mRNA表達(dá)均增強(qiáng)。提示在回轉(zhuǎn)模擬失重條件下 ,心肌成纖維細(xì)胞Ⅰ型膠原蛋白沉積增加 ,作為膠原降解抑制物的TIMP可能是造成膠原沉積的原因
