衛(wèi)星搭載藥用植物曼陀羅遺傳變異的隨機擴增多態(tài)性DNA分析

【摘要】:目的 以曼陀羅種子為材料探討太空環(huán)境對藥用植物的影響。方法 利用返回式衛(wèi)星搭載曼陀羅的種子 ,返回地面后播于實驗田中 ,應(yīng)用隨機擴增多態(tài)性 DNA(RAPD)分子標記技術(shù)分析不同組曼陀羅基因組的變異情況。結(jié)果 從 6 5個供試引物中篩選出 15個能夠產(chǎn)生可重復(fù)多態(tài)性擴增產(chǎn)物的引物。與地面對照組相比 ,失重組共產(chǎn)生 39條多態(tài)性片段 ,基因組的多態(tài)性頻率為 2 3.1% ;擊中組共產(chǎn)生 45條多態(tài)性片段 ,基因組的多態(tài)性頻率為 2 4.4%。多態(tài)性片段大小在 2 0 0~ 190 0 bp之間。結(jié)論 微重力引起曼陀羅一定程度的基因組變異 ,微重力和高能離子輻射的復(fù)合效應(yīng)大于微重力本身對植物的影響。
[Abstract]:Objective to study the effect of space environment on medicinal plants with the seeds of Datura. Methods the seeds of mandala were carried on a recoverable satellite and then returned to the ground and sown in the experimental field. Random amplified polymorphic DNA (RAPD) molecular markers were used to analyze the variation of the genome of different groups of mandola. Results Fifteen primers were screened out of 65 primers to produce repeatable polymorphic amplification products. Compared with the ground control group, 39 polymorphic fragments were produced, the frequency of genomic polymorphism was 23. 1%, and 45 polymorphic fragments were produced in the hit group, and the polymorphism frequency of the genome was 24. 4%. The polymorphic fragment size ranged from 200 BP to 1900 BP. Conclusion microgravity induces a certain degree of genomic variation in Datura, and the compound effect of microgravity and high energy ion radiation is greater than that of microgravity itself on plants.
