噪聲性聽覺損傷易感豚鼠的差異表達基因篩選研究

【摘要】: 噪聲性聽覺損傷(NIHL)是軍事噪聲環(huán)境下官兵常發(fā)生的一種聽力疾患。本研究觀察了不同噪聲暴露量對豚鼠聽覺功能和聽覺器官結(jié)構(gòu)的影響,探討了NIHL易感性研究的最佳噪聲暴露方案,建立了NIHL易感豚鼠模型,利用抑制性消減雜交技術(shù)篩選NIHL易感與不易感豚鼠耳蝸的差異表達基因片段148條,生物信息學結(jié)果顯示同源性大于80%且匹配堿基大于100 bp的片段共34條。初步分析提示線粒體基因組和熱休克蛋白與NIHL及其易感性的發(fā)生相關(guān)。研究結(jié)果將為進一步探討NIHL易感性的發(fā)生機制提供基礎。
[Abstract]:Noise-induced hearing impairment (NIHL) is a common hearing disorder in military noise environment. In this study, the effects of different noise exposures on auditory function and auditory organ structure in guinea pigs were observed, the optimal noise exposure scheme for the study of NIHL susceptibility was discussed, and the NIHL susceptible guinea pig model was established. Suppression subtractive hybridization was used to screen 148 differentially expressed genes from the cochlea of susceptible and unsusceptible guinea pigs. Bioinformatics showed that there were 34 fragments with homology of more than 80% and matching base of more than 100bp. Preliminary analysis indicated that mitochondrial genome and heat shock protein were associated with NIHL and its susceptibility. The results will provide a basis for further study on the pathogenesis of NIHL susceptibility.
