【摘要】：目的在以表面等離子體共振生物傳感器為核心的便攜式生物分子在線分析系統(tǒng)的基礎(chǔ)上,建立一種小分子檢測(cè)方法,滿足航天員體液在線檢測(cè)需求。方法建立小分子物質(zhì)2,4二硝基苯肼(DNPH)的檢測(cè)模型。用DNP BSA制備生物分子膜;采用抑制檢測(cè)法,得到檢測(cè)區(qū)間及標(biāo)準(zhǔn)曲線;對(duì)檢測(cè)的可重復(fù)性和特異性進(jìn)行了實(shí)驗(yàn)。結(jié)果1)DNPH檢測(cè)的線性范圍為7.8ng/ml～2μg/ml,其檢測(cè)下限為2.5ng/ml;2)生物分子膜的制備及傳感器的再生方法能滿足多樣品測(cè)定的需求。結(jié)論本方法靈敏、快速、簡(jiǎn)捷地實(shí)現(xiàn)了小分子物質(zhì)檢測(cè),且重復(fù)性好、可信度高,在航天醫(yī)學(xué)檢測(cè)中具有廣泛的應(yīng)用前景。
[Abstract]:Aim based on the portable on-line analysis system of biomolecules based on the surface plasmon resonance biosensor, a small molecular detection method was established to meet the requirements of the on-line detection of astronauts' body fluids. Methods the detection model of DNPH was established for the small molecular substance 2o 4 dinitrophenylhydrazide (DNPH). The biomolecular membrane was prepared by DNP BSA, the detection interval and standard curve were obtained by inhibition detection method, and the repeatability and specificity of the detection were tested. Results the linear range of 1)DNPH detection was 7.8ng/ml~2 渭 g / ml, the detection limit was 2.5 ng / ml ~ (-2) the preparation of biomolecular membrane and the regeneration method of biosensor could meet the needs of multi-sample determination. Conclusion the method is sensitive, rapid and simple for the detection of small molecular matter, and has good reproducibility and high reliability. It has a wide application prospect in space medicine detection.
