【摘要】：目的觀察成熟耳石破壞再生過程中,Otoconin 90(OC90)蛋白在內(nèi)耳的分布表達(dá)情況,探討其在成熟耳石破壞后修復(fù)再生過程中的意義。方法采用機(jī)械過載作用的方法造成豚鼠耳石破壞模型,免疫組織化學(xué)檢測(cè)耳石破壞后不同時(shí)期(0.5～1 h,3 d,7 d)內(nèi)耳切片各主要部位OC90分布表達(dá)情況,并測(cè)定橢圓囊囊斑的光密度(OD)值,對(duì)其表達(dá)強(qiáng)度進(jìn)行半定量分析。結(jié)果 OC90蛋白的分布在耳石破壞后沒有顯著變化,仍呈現(xiàn)"多點(diǎn)"表達(dá)的特點(diǎn),其在橢圓囊和球囊囊斑感覺上皮的表達(dá)于破壞后呈現(xiàn)增強(qiáng)趨勢(shì),其中耳石破壞3 d時(shí)的OD值增加顯著(P㩳0.05)。結(jié)論 OC90蛋白在成熟耳石破壞后修復(fù)再生過程中起重要作用。
[Abstract]:Objective to observe the distribution and expression of Otoconin 90 OC90 (Otoconin 90 OC90) protein in the inner ear during the regeneration of mature otolith and to explore its significance in the repair and regeneration of mature otolith. Methods the damage model of guinea pig otolithiasis was established by mechanical overload. The distribution of OC90 was detected by immunohistochemistry in the main parts of the inner ear sections at different stages after the destruction. The optical density of the oval bursa was measured. The expression intensity was analyzed by semi-quantitative analysis. Results there was no significant change in the distribution of OC90 protein after otolithiasis, and the expression of OC90 protein still showed the characteristics of "multi-point" expression. The expression of OC90 protein in the sensory epithelium of elliptical sac and balloon plaque showed an increasing trend after destruction. The OD value of otolith was significantly increased after 3 days of damage. Conclusion OC90 protein plays an important role in the repair and regeneration of mature otolith.
