【摘要】：目的探討氯化膽堿對尾吊大鼠比目魚肌肌萎縮的影響。方法按體重配對原則將24只雌性SD大鼠分為對照組(CON)、尾吊組(TS)與尾吊給藥組(TS+Cch),每組8只。尾部懸吊法建立模擬失重模型,1/2尾吊大鼠按150mg/kg的劑量用氯化膽堿灌胃,連續(xù)14d。Ca2+-ATPase法測定比目魚肌(SOL)的mATPase活性,常規(guī)方法制作石蠟組織切片。結(jié)果1)以氯化膽堿灌胃后,尾吊大鼠SOL中Ⅰ型肌纖維比例明顯升高,Ⅱ型肌纖維比例明顯降低,P0.001;2)以氯化膽堿灌胃后,尾吊大鼠SOL的肌束之間間隙減小,Ⅰ型肌纖維橫截面積(CSA)明顯升高,P0.001,Ⅱ型肌纖維CSA和單根肌纖維的平均CSA亦明顯高于尾吊組,P0.05;3)以氯化膽堿灌胃后,尾吊大鼠SOL的梭內(nèi)肌纖維中,一核鏈纖維的mATPase染色呈陰性,其它各纖維與尾吊組相同,均呈陽性。結(jié)論氯化膽堿對模擬失重條件下大鼠SOL慢肌向快肌的轉(zhuǎn)化與模擬失重引起的肌萎縮均有顯著的對抗作用。
[Abstract]:Objective to investigate the effect of choline chloride on soleus muscle atrophy in tail suspension rats. Methods Twenty-four female Sprague-Dawley rats were divided into two groups according to the principle of body weight pairing: control group (Conn), tail suspension group (TSS-TS) and tail suspension group (8 rats in each group). The simulated weightlessness model was established by tail suspension. The rats were fed with choline chloride according to the dose of 150mg/kg. The mATPase activity of soleus muscle was determined by continuous 14d.Ca2 -ATPase method. The paraffin tissue sections were made by routine method.
