雌激素對模擬失重及飛船艙內(nèi)噪聲下豚鼠內(nèi)耳Caspase-3表達的影響

【摘要】:目的探討失重及飛船艙內(nèi)噪聲復(fù)合因素對豚鼠聽功能及內(nèi)耳凋亡標(biāo)志物Caspase-3表達的影響,并觀察雌激素對上述復(fù)合因素暴露后豚鼠聽功能損傷及內(nèi)耳細胞凋亡的防治作用。方法 36只豚鼠隨機分為4組,正常對照組6只(對照組),失重+噪聲組(A組)、雌激素治療組(B組)、雌激素預(yù)防組(C組)各10只。各實驗組動物均后肢懸吊模擬失重,同時模擬飛船艙內(nèi)噪聲暴露5天,B組暴露過程中每天肌注苯甲酸雌二醇0.08mg/kg,首劑加倍,至暴露結(jié)束后3天;C組暴露前3天開始肌注苯甲酸雌二醇0.08mg/kg,首劑加倍,持續(xù)3天。對照組不給任何處理。于實驗前、暴露后即刻和恢復(fù)3天時行聽性腦干反應(yīng)測試,免疫組織化學(xué)染色方法觀察凋亡標(biāo)志物Caspase-3在內(nèi)耳毛細胞、血管紋及螺旋神經(jīng)節(jié)細胞上的表達。結(jié)果暴露后當(dāng)天,B組ABR反應(yīng)閾低于A組和C組(P0.05);暴露前后B組ABR閾移也低于A組和C組(P0.05);恢復(fù)前后,C組ABR閾移高于A組低于B組(P0.05)。對照組毛細胞、血管紋和螺旋神經(jīng)節(jié)Caspase-3表達均為陰性;A、B、C組在暴露后即刻、恢復(fù)3天時,毛細胞、血管紋和螺旋神經(jīng)節(jié)上Caspase-3的表達均為陽性;暴露后即刻,B組毛細胞和血管紋Caspase-3表達較弱,而C組表達較強;恢復(fù)3天后,A、B、C三組毛細胞、血管紋和螺旋神經(jīng)節(jié)Caspase-3表達仍為陽性,C組表達較弱。結(jié)論使用雌激素治療可有效減少失重和噪聲復(fù)合作用暴露后即刻的聽力損失和內(nèi)耳細胞凋亡;而預(yù)防性使用雌激素對暴露后即刻的聽力損傷無保護作用,但在恢復(fù)過程中可促進其恢復(fù),減少內(nèi)耳細胞凋亡。
[Abstract]:Objective to investigate the effects of weightlessness and noise in the cabin on auditory function and Caspase-3 expression of apoptotic markers in inner ear of guinea pigs. To observe the protective effect of estrogen on auditory function injury and inner ear cell apoptosis in guinea pigs after exposure to the above complex factors, 36 guinea pigs were randomly divided into 4 groups. Six normal control groups (control group, weightlessness noise group, group A, estrogen treatment group, estrogen prevention group, group C), 10 rats in each group. At the same time, during the 5 days of noise exposure in flight cabin, estradiol benzoate was injected intramuscularly into group B (0.08 mg / kg), the first dose was doubled, and the first dose was doubled 3 days before exposure to Estradiol benzoate (0.08 mg / kg) in group C 3 days after exposure. After 3 days of treatment, the control group was given no treatment. Auditory brainstem response was measured immediately after exposure and 3 days after recovery. The apoptotic marker Caspase-3 was observed in hair cells of inner ear by immunohistochemical staining. Results the ABR response threshold of group B was lower than that of group A and group C on the day of exposure, the threshold shift of ABR in group B was also lower than that in group A and group C before and after exposure, and the threshold shift of ABR in group C was higher than that in group A before and after recovery. Group B, P0.05. the control group, hair cells, The expression of Caspase-3 in hair cell, stria vascularis and spiral ganglion were all positive in group C with negative expression of Caspase-3 in stria vascularis and spiral ganglion immediately after exposure, but Caspase-3 expression in hair cell and stria vascularis was weak in group B immediately after exposure. After 3 days of recovery, the hair cells in group C were significantly higher than those in group C, and that in group C was higher than that in group C (P < 0.05). The expression of Caspase-3 in stria vascularis and spiral ganglion was still weak in group C. Conclusion estrogen therapy can effectively reduce hearing loss and apoptosis of inner ear cells immediately after exposure to combined effects of weightlessness and noise. However, prophylactic use of estrogen had no protective effect on hearing loss immediately after exposure, but it could promote the recovery and decrease the apoptosis of inner ear cells during the recovery process.
