幾株硝化菌的篩選及其初步鑒定

【摘要】:目的 :篩選、分離能夠用于治理氨氮污染和軍事坑道生活污物、污水的硝化菌。方法 :通過富集培養(yǎng)從土壤中篩選的硝化菌 ,在進行形態(tài)學檢驗和硝化速率測定后 ,設計引物擴增硝化菌的特征性基因NorB ,隨機選擇一個PCR產(chǎn)物進行測序 ,并與Genbank上的序列比較同源性。結果 :篩選到 7株能夠降解亞硝酸鹽的細菌 ,經(jīng)革蘭氏染色后發(fā)現(xiàn)全部為陰性 ,其菌落形態(tài)以小、圓、淡黃色為主 ,細菌形態(tài)多為短桿狀 ;各菌株的硝化速率在 4 .35~ 4 .80 (mg·L 1·d 1)之間。用PCR均能擴增出預期大小片段。對其中的某一個PCR產(chǎn)物直接測序 ,與Genbank中的序列比較 ,發(fā)現(xiàn)該基因與硝化菌N .hamburgensis的Norb基因同源性高達 99%。結論 :可以初步斷定 ,我們所篩選到的菌為硝化菌
[Abstract]:Objective: to screen and isolate nitrifying bacteria which can be used to control ammonia nitrogen pollution, military tunnel sewage and sewage. Methods: after morphological examination and nitrification rate determination, the nitrifying bacteria selected from soil were enriched and cultured. Primers were designed to amplify the characteristic gene NorB of nitrifying bacteria, and a PCR product was randomly selected for sequencing. Results: seven strains of bacteria capable of degrading nitrite were screened. It was found that all of them were negative by Gram staining. The colony morphology was small, round and light yellow, and most of the bacteria were short rod shape. The nitrification rate of each strain ranged from 4.35 to 4.80 mg 路L 1 路d 1. The expected size fragments could be amplified by PCR. One of the PCR products was sequenced directly and compared with the sequence in Genbank. The homology of this gene with the Norb gene of nitrifying bacteria N. hamburgensis is as high as 99%. Conclusion: it can be preliminarily concluded that the bacteria we screened are nitrifying bacteria.
