兔坐骨神經(jīng)火器傷后腰段脊髓脂質(zhì)過氧化反應(yīng)及意義

【摘要】:目的 探討脂質(zhì)過氧化反應(yīng)在兔坐骨神經(jīng)火器傷后腰段脊髓細(xì)胞凋亡中的意義。方法 火器傷組靶點(diǎn)為兔右后肢外側(cè)坐骨神經(jīng)體表投影線中點(diǎn) ;切割傷組在同一致傷水平切斷坐骨神經(jīng)。用DNA電泳、TUNEL染色技術(shù)對(duì)腰段脊髓細(xì)胞進(jìn)行凋亡定性檢測(cè) ,用流式細(xì)胞術(shù)進(jìn)行定量檢測(cè)。同時(shí)測(cè)腰段脊髓超氧化物歧化酶 (SOD)活性和丙二醛 (MDA)含量。結(jié)果 火器傷組 1周和 2周時(shí) ,腰段脊髓神經(jīng)元和膠質(zhì)細(xì)胞發(fā)生凋亡 ,切割傷組僅在 4周時(shí)有少量運(yùn)動(dòng)神經(jīng)元發(fā)生凋亡;鹌鱾M 1d時(shí) ,MDA含量顯著下降 (P 0 0 1 ) ,3d、1周、2周時(shí) ,MDA含量顯著升高 (P 0 0 1 ) ,切割傷組MDA含量變化不明顯。結(jié)論 火器傷組細(xì)胞凋亡發(fā)生時(shí)間早、數(shù)量多 ,脂質(zhì)過氧化反應(yīng)是其發(fā)生的原因之一
[Abstract]:Objective to investigate the significance of lipid peroxidation in the apoptosis of lumbar spinal cord cells after sciatic nerve firearm injury in rabbits. Methods the target of firearm injury group was the midpoint of the projection line of the lateral sciatic nerve in the right hind limb. The sciatic nerve was transected at the same injury level in the incised group. The apoptosis of lumbar spinal cord cells was detected qualitatively by DNA electrophoresis and Tunel staining. The activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) in lumbar spinal cord were measured by flow cytometry. Results the neurons and glial cells of lumbar spinal cord were apoptotic at 1 and 2 weeks after firearm injury. In the incised group, only a few motor neurons apoptosis occurred at the 4th week, and the MDA content in the firearm injury group decreased significantly on the 1st day (P 0 01), and the MDA content increased significantly at 1 week and 2 weeks after the firearm injury (P < 0 01). Conclusion the time of apoptosis in firearm injury group is early and the quantity of apoptosis is high. Lipid peroxidation is one of the reasons for the change of MDA content in cut injury group.
