規(guī)律運(yùn)動(dòng)對(duì)增齡大鼠比目魚肌細(xì)胞凋亡與自噬及PGC-1α信號(hào)調(diào)控的影響

【摘要】:目的:探討規(guī)律性運(yùn)動(dòng)對(duì)骨骼肌肌纖維增齡性老化過程中的細(xì)胞凋亡與細(xì)胞自噬及PGC-1α信號(hào)調(diào)控的作用機(jī)制。方法:選用SPF級(jí)健康雄性3月齡(青年,n=20)、13月齡(中年,n=24)和22月齡(老年,n=24)大鼠,按體重隨機(jī)分為青年對(duì)照組(Y-SED)、青年運(yùn)動(dòng)組(Y-EX);中年對(duì)照組(M-SED)、中年運(yùn)動(dòng)組(M-EX);老年對(duì)照組(O-SED)和老年運(yùn)動(dòng)組(O-EX)。對(duì)照組3組靜息,運(yùn)動(dòng)組3組實(shí)施10周規(guī)律的遞增負(fù)荷中等強(qiáng)度有氧運(yùn)動(dòng)。采用HE染色檢測(cè)肌纖維縱橫兩個(gè)面的形態(tài)學(xué)變化,免疫印跡法檢測(cè)SOD表達(dá)水平、TUNEL法檢測(cè)凋亡,免疫印跡法等檢測(cè)自噬及PGC-1α通路的蛋白質(zhì)表達(dá)水平。結(jié)果:(1)HE染色顯示規(guī)律有氧運(yùn)動(dòng)提高了增齡性大鼠比目魚肌肌纖維的成束性和緊密性,而免疫印跡結(jié)果顯示其提高了SOD表達(dá)水平。(2)各年齡對(duì)照組大鼠比目魚肌細(xì)胞凋亡的增加呈現(xiàn)增齡性趨勢(shì),而規(guī)律有氧運(yùn)動(dòng)各年齡大鼠凋亡指數(shù)分別增加了7.55%、20.26%(Ρ0.05)和14.52%(Ρ0.05)。(3)各年齡對(duì)照組大鼠自噬基因LC-Ⅲ呈現(xiàn)增齡性降低趨勢(shì),規(guī)律有氧運(yùn)動(dòng)各年齡大鼠的LC-Ⅲ均顯著上升(Ρ0.01),各年齡運(yùn)動(dòng)組自身LC-Ⅲ卻呈現(xiàn)增齡性上升趨勢(shì)。(4)與相對(duì)應(yīng)的對(duì)照組相比較,規(guī)律有氧運(yùn)動(dòng)各年齡大鼠自噬因子Beclin1表達(dá)均顯著上升(Ρ0.05),其中Y-EX組上升最顯著(Ρ0.01),各年齡運(yùn)動(dòng)組自身Be-clin1的表達(dá)水平卻呈現(xiàn)增齡性降低趨勢(shì)。(5)與Y-SED組相比,M-SED組的PGC-1α表達(dá)水平增加,但O-SED組比Y-SED和M-SED兩組的PGC-1α表達(dá)水平顯著下降(Ρ0.01)。與安靜對(duì)照組相比較,Y-EX和M-EX兩組PGC-1α表達(dá)水平均呈現(xiàn)上升趨勢(shì),其中與O-SED組相比,O-EX組PGC-1α表達(dá)水平顯著上升(Ρ0.01,Ρ0.05)。結(jié)論:自噬與凋亡兩者之間的平衡穩(wěn)定關(guān)系影響著比目魚肌增齡性老化的發(fā)生發(fā)展,規(guī)律有氧運(yùn)動(dòng)對(duì)增齡性老化大鼠比目魚肌細(xì)胞自噬與凋亡的影響出現(xiàn)增齡性變化,其機(jī)制是通過激發(fā)Beclin1和PGC-1α信號(hào)通路參與調(diào)控而達(dá)到調(diào)節(jié)比目魚肌的生物學(xué)功能與改善骨骼肌老化。
