【摘要】：目的探討低劑量氚照射對大鼠海馬神經(jīng)細(xì)胞黏附分子L1、神經(jīng)細(xì)胞黏附分子(NCAM)表達(dá)的影響。方法取24 h新生鼠腦海馬細(xì)胞培養(yǎng),培養(yǎng)第7天以3.7×102、3.7×103、3.7×104、3.7×105、3.7×106Bq/ml終濃度氚水照射細(xì)胞24 h,并設(shè)對照組(0 Bq/ml),活細(xì)胞工作站測定各處理組細(xì)胞的遷移距離;Western blot法、免疫細(xì)胞化學(xué)法檢測各濃度氚水照射下神經(jīng)細(xì)胞黏附分子L1及NCAM蛋白的表達(dá)情況。結(jié)果神經(jīng)細(xì)胞遷移距離測定結(jié)果顯示,與對照組相比,受照組細(xì)胞遷移距離明顯縮短,且隨著氚水濃度的不斷增加而逐漸縮短(P0.05);Western blot法、免疫細(xì)胞化學(xué)法檢測顯示受照組神經(jīng)細(xì)胞黏附分子L1、NCAM蛋白表達(dá)含量均低于對照組,并隨氚水濃度的升高表達(dá)量呈遞減趨勢(P0.05)。結(jié)論氚照射可致神經(jīng)細(xì)胞黏附分子表達(dá)量減少進(jìn)而可影響神經(jīng)細(xì)胞的正常遷移。
[Abstract]:Aim to investigate the effects of low dose tritium irradiation on the expression of nerve cell adhesion molecule L1 and neural cell adhesion molecule (NCAM) in hippocampus of rats. Methods the cells were cultured for 24 h and irradiated with 3.7 脳 102,3.7 脳 103,3.7 脳 104,3.7 脳 105,3.7 脳 106Bq/ml final concentration of tritium water for 24 hours. The control group (0 Bq/ml) was set up. The migration distance of cells in each treatment group was measured by living cell workstation. Western blot method and immunocytochemical method were used to detect the expression of nerve cell adhesion molecule L1 and NCAM protein under various concentrations of tritiated water irradiation. Results compared with the control group, the migration distance of neurons in the irradiated group was significantly shorter than that in the control group, and gradually shortened with the increasing of the concentration of tritium water (P0.05). Western blot assay and immunocytochemistry showed that the expression of NCAM protein and L1 in the exposed group were lower than those in the control group, and the expression level decreased with the increase of tritium water concentration (P0.05). Conclusion tritium irradiation can decrease the expression of nerve cell adhesion molecules and then affect the normal migration of neural cells.
