人白細胞中SIRT1和FoxO3a mRNA表達的隨齡變化研究

【摘要】:研究背景在目前的法醫(yī)學實踐中,個體年齡的推斷主要是根據(jù)骨骼、牙齒等骨性指標的隨齡變化特征,應(yīng)用法醫(yī)人類學模型計算來完成的。該技術(shù)已經(jīng)比較成熟,且誤差也相對較小。但在遇到骨骼、牙齒等檢材缺失的案例時,其應(yīng)用就受到了極大的限制。隨著分子生物學的發(fā)展,通過檢測端粒長度、線粒體DNA損傷程度及DNA甲基化水平等推斷個體年齡已成為研究熱點,但因種種原因,目前尚難應(yīng)用于法醫(yī)學實踐。近年來研究發(fā)現(xiàn),沉默信息調(diào)節(jié)蛋白1(SIRT1)和叉頭蛋白O亞家族3a(FoxO3a)在細胞衰老和人體長壽中發(fā)揮重要作用,這也提示SIRT1和FoxO3a的表達可能與年齡存在密切關(guān)系。因此如果能建立SIRT1和FoxO3a基因表達與年齡之間的數(shù)學模型,將有可能實現(xiàn)通過這兩種基因表達的豐度測定來推斷個體年齡,有望為法醫(yī)學中的年齡推斷提供新的技術(shù)手段。 目的通過檢測比較不同年齡段個體SIRT1和FoxO3a mRNA的表達水平,并分析其隨齡變化趨勢,為進一步建立SIRT1和FoxO3a mRNA表達與年齡之間的數(shù)學模型奠定基礎(chǔ)。 方法采集80個健康無關(guān)個體血樣,按年齡分為8組:≤9歲、10~19歲、20~29歲、30~39歲、40~49歲、50~59歲、60~69歲及≥70歲組,每組10人。提取總RNA后,利用實時定量逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(RT-qPCR)技術(shù),檢測人白細胞中SIRT1和FoxO3a mRNA的表達情況,,用SPSS軟件對數(shù)據(jù)資料進行統(tǒng)計分析。 結(jié)果⑴人白細胞SIRT1mRNA相對表達水平在≤9歲、10~19歲、20~29歲、30~39歲、40~49歲和50~59歲組之間均無顯著性差異(P>0.05),而60~69歲和≥70歲組分別與上述各組有顯著性差異(P<0.01),且此兩組之間也有顯著性差異(P<0.01)。白細胞SIRT1mRNA相對表達水平與年齡正相關(guān)(r=0.708,P<0.01),以年齡分組為自變量,SIRT1mRNA相對表達水平為因變量,進行曲線擬合得Y=0.005X3-0.002X2-0.004(R2=0.884,P<0.01);⑵人白細胞FoxO3a mRNA在≤9歲與10~19歲組之間、≤9歲與20~29歲組之間,10~19歲與20~29歲組之間,20~29歲與30~39歲組之間,30~39歲與40~49歲組之間相對表達水平無顯著性差異(P>0.05),其余任何兩組之間白細胞FoxO3a mRNA相對表達水平有顯著性差異(P<0.05)。白細胞FoxO3amRNA相對表達水平與年齡正相關(guān)(r=0.841, P<0.01),以年齡分組為自變量,F(xiàn)oxO3a mRNA相對表達水平為因變量,進行曲線擬合得Y=0.001e0.418x(R2=0.935,P<0.01)。 結(jié)論⑴人白細胞SIRT1mRNA在健康個體年齡㩳60歲時表達量非常低,而在≥60歲時高表達;⑵不同年齡組健康個體白細胞FoxO3a mRNA表達隨年齡的增長而升高;⑶人白細胞FoxO3a mRNA表達水平有望成為年齡推斷的有用指標之一。
[Abstract]:Background in the current forensic practice, the estimation of individual age is mainly based on the characteristics of bone, teeth and other bone indicators with age, using forensic anthropology model to complete the calculation. The technology has been more mature, and the error is relatively small. However, in the case of missing bone, teeth and other materials, its application is greatly limited. With the development of molecular biology, the estimation of individual age by detecting telomere length, mitochondrial DNA damage and DNA methylation has become a hot topic. However, it is difficult to apply it to forensic practice for various reasons. In recent years, silencing information regulatory protein 1 (SIRT1) and fork head protein O subfamily 3a (FoxO3a) have been found to play an important role in cell senescence and longevity, which suggests that the expression of SIRT1 and FoxO3a may be closely related to age. Therefore, if a mathematical model between SIRT1 and FoxO3a gene expression and age can be established, it will be possible to infer individual age by determining the abundance of these two genes, which may provide a new technique for age estimation in forensic medicine. Objective to compare the expression levels of SIRT1 and FoxO3a mRNA in individuals of different ages and analyze their trends with age so as to establish a mathematical model between SIRT1 and FoxO3a mRNA expression and age.
