有氧和抗阻運動對大鼠白色脂肪棕色化的作用

【摘要】:目的:對比兩種不同的運動方式——有氧運動和抗阻運動對大鼠白色脂肪棕色化的作用。方法:雄性SD大鼠隨機分為3組,分別為安靜對照組(C)、有氧訓練組(T)和抗阻訓練組(L),有氧訓練方式為跑臺訓練:8周的時間里速度由15 m/min增至28 m/min,訓練時間由20 min增至60 min,每周訓練5天。抗阻訓練方式為遞增負荷爬梯訓練,爬梯長1.1 m,負重裝置固定于大鼠尾部,初始負荷為體重的50%,之后不斷增加負荷,每3天進行1次,共訓練8周。最后一次訓練結束后48 h取大鼠的雙側腹股溝脂肪和附睪脂肪,組織進行HE染色,熒光定量PCR,觀察兩種運動對脂肪組織形態(tài)和相關基因mRNA的影響。結果:T組腹股溝脂肪和附睪脂肪的脂滴面積都顯著減小,腹股溝脂肪組織里還出現(xiàn)了一些多腔室的小細胞成簇的聚集和少量類似血管的管狀物;L組兩類脂肪的脂滴面積比C組略有減小但不顯著;T組的PRDM16、PPARγ、PGC-1α、UCP1的mRNA在腹股溝脂肪中的表達量與C組相比有顯著升高,在附睪脂肪中的表達量除PPARγ以外都略有降低,L組的上述基因在腹股溝脂肪中表達量與C組相比略有升高但無顯著性,在附睪脂肪中的表達量也表現(xiàn)為除PPARγ以外都出現(xiàn)不同程度的降低。結論:有氧運動可以促進皮下白色脂肪棕色化,但對內臟脂肪作用不明顯;抗阻運動對皮下和內臟脂肪均無促進其發(fā)生棕色化的作用。
[Abstract]:Aim: to compare the effects of aerobic exercise and resistance exercise on white fat browning in rats. Methods: male SD rats were randomly divided into three groups: quiet control group, (C), aerobic training group (T) and resistance training group (L), aerobic training method: the speed increased from 15 m/min to 28 m / min in 8 weeks. The training time was increased from 20 min to 60 min, per week. The resistance training method was incremental load climbing training, the climbing ladder length was 1.1 m, the load loading device was fixed on the tail of the rats, the initial load was 50 times of body weight, and then the load was continuously increased, once every 3 days, for a total of 8 weeks. The bilateral inguinal fat and epididymal fat of the rats were taken 48 hours after the last training and the tissues were stained with HE. The effects of the two exercises on the morphology of adipose tissue and the related gene mRNA were observed by fluorescence quantitative PCR,. Results: in group T, the area of fat droplets in inguinal fat and epididymal fat decreased significantly, and there were some clusters of small cells in groin adipose tissue and a small number of vascular-like tubules. The lipid drop area of group L was slightly smaller than that of group C, but it was not significant. The expression of PRDM16,PPAR 緯, PGC-1 偽 and UCP1 in inguinal fat in T group was significantly higher than that in C group, and the expression of PPAR 緯 in epididymal fat was slightly decreased. The expression of these genes in the inguinal fat of group L was slightly higher than that in group C, but the expression level in the epididymal fat was also decreased in varying degrees except PPAR 緯. Conclusion: aerobic exercise can promote the browning of subcutaneous white fat, but it has no obvious effect on visceral fat, and resistance exercise has no effect on the browning of subcutaneous and visceral fat.
