【摘要】：目的研究刺梨黃酮(FRT)對(duì)輻射損傷骨髓細(xì)胞周期的影響。方法取小鼠骨髓細(xì)胞制成單細(xì)胞懸液,分為空白對(duì)照組、輻射組、輻射+50 mg·L~(-1)FRT干預(yù)組(50 mg·L~(-1)FRT干預(yù)組)、輻射+100 mg·L~(-1)FRT干預(yù)組(100 mg·L~(-1)FRT干預(yù)組)�？瞻讓�(duì)照組不加干預(yù);輻射組給予6 Gyγ射線照射;50、100 mg·L~(-1)FRT干預(yù)組先在骨髓細(xì)胞懸液加入相應(yīng)量的FRT,使FRT終濃度分別為50、100 mg·L~(-1),2 h后給予6 Gyγ射線照射。經(jīng)細(xì)胞固定、碘化丙啶染色后,進(jìn)行流式細(xì)胞儀檢測(cè)。結(jié)果與空白對(duì)照組比較,輻射組照射后6、12 h,G_1期、S期細(xì)胞比例均降低(P0.05),G_2期細(xì)胞比例均增高(P0.05),且輻射后12 h G_1和S期細(xì)胞比例低于6 h時(shí)(P0.05),G_2期細(xì)胞比例顯著高于6 h時(shí)(P0.05)。與輻射組比較,50、100 mg·L~(-1)FRT干預(yù)組的G_1、S期細(xì)胞比例均增高(P0.05),G_2期細(xì)胞比例均減少(P0.05),且輻射后6、12 h,100 mg·L~(-1)FRT干預(yù)組G_1和S期細(xì)胞比例高于50 mg·L~(-1)FRT干預(yù)組(P0.05),G_2期細(xì)胞比例低于50 mg·L~(-1)FRT干預(yù)組(P0.05)。結(jié)論 FRT可以降低輻射后骨髓細(xì)胞G_2期的細(xì)胞比例及增高G_1、S期的細(xì)胞所占比例,對(duì)γ射線所致骨髓細(xì)胞損傷有一定防護(hù)作用,且在一定濃度范圍內(nèi)防護(hù)效果呈濃度依賴性。
[Abstract]:Objective to study the effect of Rosa roxburghii flavonoids (FRT) on bone marrow cell cycle induced by radiation injury. Methods Mouse bone marrow cells were made into single cell suspension and divided into control group, 50 mg L ~ (-1) FRT intervention group (50 mg L ~ (-1) FRT intervention group) and 100 mg L ~ (-1) FRT intervention group (100 mg L ~ (-1) FRT intervention group). The control group was treated with 6 Gy 緯 -ray irradiation for 50 mg L ~ (-1) FRT, and the bone marrow cell suspension was exposed to the corresponding amount of FRT, for 2 h after the final concentration of FRT was 50100 mg L ~ (-1) ~ (-1) for 2 h. The cells were fixed and stained with pyridine iodide, and detected by flow cytometry. Results compared with the control group, the proportion of S phase cells in the radiation group decreased (P0.05) and the proportion of the cells in the GSP 2 phase increased significantly (P0.05), and at 12 h after irradiation, the proportion of the cells in the G 1 and S phase was lower than that in the 6 h (P0.05). The percentage of the cells in the G 2 phase was significantly higher than that in the 6 h group (P0.05). Compared with the radiation group, the proportion of G1S phase cells in the 50 100 mg L ~ (-1) FRT group was increased (P0.05), and the proportion of G1 and S phase cells in the G1 and S phase group was higher than that in the 50 mg L ~ (-1) FRT intervention group (P0.05). The percentage of G1 and S phase cells in the G1 and S phase group was higher than that in the 50 mg L ~ (-1) FRT intervention group (P0.05). The percentage of G1 and S phase cells in the G1 and S phase group was significantly higher than that in the 50 mg L ~ (-1) FRT intervention group (P0.05). Conclusion FRT can decrease the proportion of bone marrow cells in Gast2 phase after irradiation and increase the proportion of cells in G _ 1 / S phase. It has a protective effect on bone marrow cell injury induced by 緯 -ray, and the protective effect is concentration-dependent within a certain concentration range.
