運(yùn)動(dòng)對(duì)成長(zhǎng)期骨骼肌神經(jīng)源性誘導(dǎo)因子Agrin及其受體MuSK表達(dá)的影響

【摘要】:目的:本研究通過(guò)分析神經(jīng)源性誘導(dǎo)因子Agrin及其受體-肌特異性受體酪氨酸激酶Mu SK在成長(zhǎng)期(離乳-性成熟)骨骼肌的表達(dá)水平,探討其在神經(jīng)肌肉連接(NMJ)發(fā)育中的作用及早期運(yùn)動(dòng)干預(yù)的影響。方法:出生18天SD雄性大鼠130只隨機(jī)分為運(yùn)動(dòng)組50只、懸吊組和對(duì)照組各40只,運(yùn)動(dòng)組和懸吊組分別進(jìn)行運(yùn)動(dòng)增強(qiáng)(跑臺(tái)運(yùn)動(dòng))和運(yùn)動(dòng)減弱(尾部懸吊)干預(yù)。對(duì)照組3周齡時(shí),各組4周齡、5周齡、6周齡、8周齡時(shí)分別取材10只。測(cè)定并比較正常發(fā)育(3~8周齡)及運(yùn)動(dòng)干預(yù)下腓腸肌Agrin和Mu SKm RNA水平(RT-PCR法)及腓腸肌Mu SK蛋白水平(ELISA法)的差異。結(jié)果:(1)對(duì)照組的Agrin m RNA在4周齡較3周齡表達(dá)減少(P0.01),5周齡再次出現(xiàn)表達(dá)高峰(P0.01),在4周齡后Mu SK m RNA表達(dá)減少(P0.01),6周齡再次出現(xiàn)Mu SK m RNA表達(dá)高峰(P0.01);運(yùn)動(dòng)組Mu SK m RNA表達(dá)水平在5周齡時(shí)顯著高于對(duì)照組(P0.01),而懸吊組Mu SK m RNA表達(dá)水平在各周齡均低于對(duì)照組(P0.05,P0.01)。(2)運(yùn)動(dòng)組Mu SK蛋白水平在各周齡組均高于對(duì)照組(P0.05);懸吊組在4周齡時(shí)低于對(duì)照組(P0.05)。結(jié)論:成長(zhǎng)期腓腸肌Agrin及其受體Mu SK的表達(dá)具有一定的時(shí)間規(guī)律,在出生3~4周(離乳前后)呈現(xiàn)逐漸減少的趨勢(shì),5~6周(青春期)再次出現(xiàn)表達(dá)高峰;在此期間進(jìn)行適當(dāng)?shù)倪\(yùn)動(dòng)訓(xùn)練可以促使Agrin及其受體Mu SK表達(dá)增多,這可能作為一種機(jī)制,促進(jìn)NMJ在成長(zhǎng)期的發(fā)育過(guò)程。
[Abstract]:Objective: to investigate the expression of neurogenic inducible factor (Agrin) and its receptor-specific receptor tyrosine kinase (Mu SK) in adult skeletal muscle. To explore its role in the development of neuromuscular junction (NMJ) and the effect of early exercise intervention. Methods: one hundred and thirty male SD rats were randomly divided into two groups: exercise group (n = 50), suspension group (n = 40) and control group (n = 40). In the control group, 10 rats were collected at the age of 3 weeks, 4 weeks, 5 weeks, 6 weeks and 8 weeks. The levels of Agrin and Mu SKm RNA in gastrocnemius muscle (RT-PCR method) and Mu SK protein level in gastrocnemius muscle (ELISA method) were measured and compared under normal development (3 ~ 8 weeks old) and exercise intervention. Results: (1) the expression of Agrin m RNA in the control group was decreased at the age of 4 weeks compared with that in the age of 3 weeks (P0.01). The peak expression of Mu SK m RNA was again found in the control group at the age of 5 weeks (P0.01), and in the control group at the age of 4 weeks (P0. 01) and at the age of 6 weeks (P0. 01), and in the exercise group, the expression of Mu SK m RNA appeared again (P0. 01). The expression level of Mu SK m RNA in the suspension group was significantly higher than that in the control group at the age of 5 weeks (P0.01), while the Mu SK protein level in the exercise group was significantly higher than that in the control group at the age of 5 weeks (P0.05), and in the suspension group was lower than that in the control group at 4 weeks of age (P0.05). Conclusion: the expression of Agrin and its receptor Mu SK in gastrocnemius muscle has a certain time regular pattern. The expression peak of Agrin and its receptor Mu SK in gastrocnemius muscle is decreasing gradually at 3 ~ 4 weeks (before and after the birth), and it appears again at 5 ~ 6 weeks (puberty). During this period, proper exercise training can increase the expression of Agrin and its receptor Mu SK, which may be used as a mechanism to promote the development of NMJ in the process of development.
