不同強度運動對肥胖大鼠肝臟氧化應(yīng)激和脂聯(lián)素及其受體的影響

【摘要】:目的:探討三種不同強度運動對肥胖大鼠肝臟氧化應(yīng)激及脂聯(lián)素/脂聯(lián)素受體等相關(guān)指標(biāo)的影響。方法:經(jīng)16周高脂飼料喂養(yǎng)構(gòu)建肥胖大鼠模型,32只模型鼠分為高脂安靜組(HF,8只)、小強度運動組(HS組,8只)、中等強度運動組(HM組,8只)和遞增負荷運動組(HI組,8只),另外選取正常對照組(NC組)大鼠8只。運動方式采用跑臺運動,小強度運動跑速為10 m/min,中等強度跑速為15 m/min,遞增負荷強度為10 m/min的速度運動20 min、20 m/min的速度運動20 min、28~30 m/min的速度運動20min。運動組每周運動5天,每次1小時。干預(yù)持續(xù)6周后檢測各組大鼠體重,肝臟超氧化物歧化酶(SOD)活性、丙二醛(MDA)、腫瘤壞死因子α(TNF-α)、白介素6(IL-6)、脂聯(lián)素受體I(AR1)、AR2,血清谷丙轉(zhuǎn)氨酶(ALT)、谷草轉(zhuǎn)氨酶(AST)、脂聯(lián)素、甘油三酯(TG)、膽固醇(TC)、高密度脂蛋白膽固醇(HDL-C)、低密度脂蛋白膽固醇(LDL-C)、自由脂肪酸(FFA)等指標(biāo)。結(jié)果:與NC組比較,HF組大鼠體重、TG、TC、LDL-C、FFA含量、ALT活性、AST活性以及MDA、TNF-α和IL-6表達均顯著性升高(P0.05或P0.01),HDL-C含量、SOD活性、血清脂聯(lián)素水平及AR1、AR2表達均顯著性降低(P0.05或P0.01);與HF組比較,HS、HM和HI組體重、血清TG、LDL-C、FFA含量及肝臟ALT含量、MDA、TNF-α、IL-6表達均顯著性降低(P0.05或P0.01),SOD活性、脂聯(lián)素水平及AR1、AR2表達均顯著性升高(P0.05或P0.01),僅HM組HDL-C含量顯著升高(P0.05);與HS組和HI組比較,HM組FFA含量、MDA表達進一步降低(P0.05),脂聯(lián)素水平、AR2進一步增加(P0.05)。結(jié)論:有氧運動能有效改善高脂飲食所致的肥胖大鼠脂代謝異常,提高肝臟抗氧化酶活性,增強自由基清除能力,預(yù)防肝細胞炎癥損傷,改善肝臟代謝功能紊亂,其中中等強度運動優(yōu)于小強度或遞增負荷強度運動,其機制可能是通過增加血清脂聯(lián)素進一步激活肝臟脂聯(lián)素受體2有關(guān)。
[Abstract]:Aim: to investigate the effects of three different intensities of exercise on liver oxidative stress and adiponectin / adiponectin receptor in obese rats. Methods: after 16 weeks of high fat diet feeding, 32 obese rats were divided into four groups: hyperlipidemia group (8 rats), low intensity exercise group (HS group, 8 rats), moderate intensity exercise group (HM group, 8 rats) and increasing load exercise group (HI group, 8 rats). In addition, 8 normal control group (NC group) rats were selected. The treadmill movement was used, the low intensity running speed was 10 m / min, the moderate intensity running speed was 15 m / min, the incremental load intensity was 10 m/min, the speed movement was 20 mins and 20 m/min, 20 mins and 2830 m/min. Exercise group 5 days a week, 1 hour each time. Body weight, liver superoxide dismutase (SOD), malondialdehyde (MDA), tumor necrosis factor- 偽 (TNF- 偽), interleukin-6 (IL-6), adiponectin receptor I (AR1) AR2, serum alanine aminotransferase (alt), aspartate aminotransferase (AST), adiponectin were measured after 6 weeks of intervention. Triglyceride (TG), cholesterol (TC), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), free fatty acid (FFA) and so on. Results: compared with NC group, the body weight of rats in HF group was significantly increased (P0.05 or P0.01), serum adiponectin level and AR1 / AR2 expression were significantly decreased (P0.05 or P0.01), and compared with HF group, the activity of alt and the expression of MDA-TNF- 偽 and IL-6 were significantly increased (P0.05 or P0.01), and the body weight of HSHM and HI group was significantly lower than that of HF group (P0.05 or P0.01), and compared with that of HF group, the activity of alt and the expression of MDA-TNF- 偽 and IL-6 were significantly increased (P0.05 or P0.01). The levels of serum TGG LDL-CfFA and liver alt were significantly decreased (P0.05 or P0.01), and the activity of SOD was significantly decreased in MDA-TNF- 偽 and IL-6 (P0.05 or P0.01), and the expression of TNF- 偽 and IL-6 decreased significantly (P0.05 or P0.01). Adiponectin level and AR1P AR2 expression were significantly increased (P0.05 or P0.01), only HDL-C content in HM group was significantly increased (P0.05), compared with HS group and HI group, FFA content and MDA expression in HM group were further decreased (P0.05), adiponectin level and AR2 level were further increased (P0.05). Conclusion: aerobic exercise can effectively improve lipid metabolism of obese rats induced by high fat diet, increase liver antioxidant enzyme activity, enhance free radical scavenging ability, prevent hepatocyte inflammation injury and improve liver metabolic disorder. The moderate intensity exercise is superior to small intensity exercise or increasing load intensity exercise, and the mechanism may be related to the further activation of hepatic adiponectin receptor 2 by increasing serum adiponectin.
