【摘要】：為探討安多霖對(duì)微波輻射損傷的的防治機(jī)制,將SD雄性大鼠按質(zhì)量隨機(jī)分為正常對(duì)照組、輻照組和預(yù)防組(給藥量分別為3、6、9 mg/kg)。預(yù)防組大鼠連續(xù)灌胃給藥,對(duì)照組和輻照組給予雙蒸水,20 d后輻照組和預(yù)防組大鼠行100 m W/cm~2的高功率微波(High power-microwave,HPM)全身輻照10 min。于輻照后第1、2、5天取睪丸組織制備病理標(biāo)本,采用SP免疫組化檢測(cè)試劑盒標(biāo)定目的蛋白,Image-pro plus圖像分析軟件對(duì)特定染色區(qū)域進(jìn)行灰度分析和測(cè)定。結(jié)果顯示,輻照組大鼠睪丸細(xì)胞Caspase-9水平較對(duì)照組呈"高-低-高"的"倒拋物線"形(p0.05),給藥組則隨著時(shí)間呈現(xiàn)"先低后高"的趨勢(shì)(p0.05);對(duì)于X連鎖凋亡抑制蛋白(X-linked inhibitor of apoptosis protein,XIAP)而言,輻照組與對(duì)照組之間的差異不顯著,而6、9 mg/kg組在輻照后第5天較輻照組顯著升高(p0.05)。結(jié)果提示,Caspase-9參與了HPM致大鼠睪丸細(xì)胞凋亡的過(guò)程,安多霖對(duì)大鼠睪丸細(xì)胞的保護(hù)作用需要Caspase-9和XIAP的共同參與。
[Abstract]:In order to investigate the mechanism of Andolin preventing and treating microwave radiation injury, SD male rats were randomly divided into normal control group, irradiation group and prevention group according to their mass. Rats in the prevention group were given intragastric administration continuously, the control group and the irradiation group were given double-steamed water for 20 days. The rats in the irradiation group and the prevention group were exposed to 100m W/cm~2 high power microwave (HPM) for 10 mins. Testicular tissues were taken from testis 5 days after irradiation, and the specific staining regions were analyzed and measured by SP immunohistochemical detection kit calibrating the target protein image-pro plus image analysis software. The results showed that the Caspase-9 level of testicular cells in the irradiated group was "high-low-high" inverted parabola "in comparison with the control group, while in the treatment group it was" low first and then high "(p0.05), and for X-linked inhibitor of apoptosis protein (XIAPs) of X-linked inhibitor of apoptosis protein (X-linked inhibitor of apoptosis protein), the level of Caspase-9 in the irradiated group was higher than that in the control group. There was no significant difference between the irradiation group and the control group, but the 6on 9 mg/kg group was significantly higher than that of the irradiation group on the 5th day after irradiation. The results suggested that Caspase-9 was involved in the apoptosis of rat testicular cells induced by HPM, and the protective effect of Andolin on rat testicular cells required the joint participation of Caspase-9 and XIAP.
