【摘要】：目的:比較耐力運動及力竭運動對大鼠血液及肝臟中鐵調素(hepcidin)表達的影響,并測定分析與之相關的鐵代謝指標、低氧應答因子、炎性因子、抗菌肽活性相關指標的變化趨勢。方法:雌性SD大鼠30只,隨機分為對照組(n=10)、耐力運動組(n=10)、力竭運動組(n=10),對照組不運動,耐力運動組和力竭運動組分別采用為期4周的中等運動強度跑臺訓練和力竭運動強度跑臺訓練對大鼠進行干預,而后分別采集血樣,進行血常規(guī)指標及血清中鐵轉運因子、低氧應答因子、炎性因子及抗菌肽活性相關指標測定;取肝臟標本,固定、包埋、切片后,利用免疫組織化學染色分別測定不同組別動物肝臟中hepcidin、低氧誘導因子(HIF-1α)、核轉錄因子kappa b(NF-κB)含量。結果:與對照組相比,4周耐力性運動后大鼠血清中hepcidin水平降低(P0.05),促紅細胞生成素(EPO)濃度升高(P0.05),白細胞介素6(IL-6)濃度下降(P0.01)。與對照組相比,4周力竭性運動后大鼠血清及肝臟中hepcidin水平升高(P0.01),EPO濃度降低(P0.01),同時伴有血清及肝臟中HIF-1α表達增加(P0.01),血清轉鐵蛋白(TF)、可溶性轉鐵蛋白受體(s TFR)水平降低(P0.01),血清中白細胞介素1(IL-1)、IL-6、腫瘤壞死因子α(TNF-α)、C反應蛋白(CRP)水平顯著升高(P0.01,P0.001,P0.001,P0.001);同時血清中與hepcidin抗菌肽活性相關的toll樣受體4(TLR4)、NF-κB水平均顯著升高(P0.001)。結論:4周耐力性運動可下調大鼠血清中hepcidin水平;而4周力竭性運動可使大鼠血清及肝臟中hepcidin水平顯著上調,提示不同運動方式對hepcidin的影響存在明顯差異。
[Abstract]:Aim: to compare the effects of endurance exercise and exhaustive exercise on the expression of heparin hepcidin in blood and liver of rats, and to determine the related indexes of iron metabolism, hypoxia response factor, inflammatory factor and antibacterial peptide activity. Methods: thirty female Sprague-Dawley rats were randomly divided into control group (n = 10), endurance exercise group (n = 10), exhaustive exercise group (n = 10) and control group (n = 10). Rats in endurance exercise group and exhaustive exercise group were treated with moderate intensity treadmill training and exhaustive exercise intensity treadmill training for 4 weeks respectively. Blood samples were collected and blood routine indexes and iron transport factors in serum were collected. The activity of hypoxia response factor, inflammatory factor and antimicrobial peptide were measured. The contents of hepcidin, hypoxia-inducible factor (HIF-1 偽) and nuclear transcription factor (kappa) NF- 魏 B in the liver of different groups of animals were determined by immunohistochemical staining. Results: compared with the control group, the levels of serum hepcidin, erythropoietin (EPO), interleukin-6 (IL-6) and interleukin-6 (IL-6) decreased in rats after 4 weeks of endurance exercise. Compared with the control group, the level of hepcidin in serum and liver of rats after 4 weeks exhaustive exercise was higher than that of the control group. The concentration of P0.01a was decreased, the expression of HIF-1 偽 in serum and liver was increased, and the levels of serum transferrin, transferrin receptor and soluble transferrin receptor were decreased. The levels of IL-6, TNF- 偽 -TNF- 偽 -C-reactive protein (CRP-) in serum were significantly higher than those in control group, and the levels of toll receptor 4TLR4NF-魏 B related to the activity of hepcidin antimicrobial peptide in serum were significantly higher than that of P0.001, and the levels of toll receptor 4TLR4NF-魏 B in serum were significantly higher than those in the control group (P < 0.05), while the levels of TNF- 魏 B in serum were significantly higher than those in the control group (P < 0.05). Conclusion the level of hepcidin in serum can be down-regulated by endurance exercise for 4 weeks, and the level of hepcidin in serum and liver can be upregulated by exhaustive exercise at 4 weeks, indicating that there are significant differences in the effect of different exercise modes on hepcidin.
