放射性腦損傷的MRS、PWI動物實驗研究

【摘要】:目的:建立放射性腦損傷的大鼠模型,研究MRS、PWI在放射性腦損傷不同時期的改變,為放射性腦損傷的早期診斷提供依據(jù),并初步探討放射性腦病可能的發(fā)病機制。 材料與方法:將成年雌性Wistar大鼠30只隨機分為空白對照組和單純照射組,均進行MRI、MRS及MR-PWI檢查。單純照射組采用6Mev的電子線對大鼠進行20Gy單次全腦垂直照射,分別于照射后1、3、6、9及12個月行MRI、MRS及MR-PWI檢查,分析N-乙酰天門冬氨酸(NAA)、膽堿(Cho)、肌酸(Cr)等含量的變化,采用積分面積進行比較,計算NAA/Cr和Cho/Cr (?)匕值,同時在MR后處理工作站獲得PWI偽彩圖,測量灌注后大鼠雙側(cè)側(cè)腦室周圍的rCBV值。 結(jié)果:①所有被檢查大鼠的大腦常規(guī)MRI均未見異常表現(xiàn)。②各照射組大鼠的NAA/Cr比值與對照組相比隨時間延長逐漸下降,且均有統(tǒng)計學意義(P0.05);照射后Cho/Cr比值先上升,3個月時達峰值,隨后逐漸下降,6個月內(nèi)各組的Cho/Cr與對照組相比有統(tǒng)計學意義(P0.05)。③照射后1個月組大鼠的rCBV與對照組相比無明顯統(tǒng)計學意義。照射1個月后各組的rCBV值均明顯低于對照組,差異有統(tǒng)計學意義(P0.05) 結(jié)論:①MRS能夠在放射性腦損傷發(fā)生形態(tài)學改變之前,檢測出腦組織代謝的異常 ②PWI能夠在放射性腦損傷發(fā)生形態(tài)學改變之前,檢測出腦組織微循環(huán)的異常。
[Abstract]:Objective: to establish a rat model of radiation brain injury and to study the changes of MRS PWI in different stages of radiation brain injury, to provide evidence for the early diagnosis of radiation brain injury, and to explore the possible pathogenesis of radiation encephalopathy. Materials and methods: thirty adult female Wistar rats were randomly divided into blank control group and irradiation group. MRI Mrs and MR-PWI were performed. Rats in the single irradiation group were exposed to 6Mev single whole brain vertical irradiation. MRIMRS and MR-PWI were performed at 1 and 12 months after irradiation to analyze the changes of the contents of N-acetylaspartate, choline chorion and creatine. The NAA/Cr and Cho/Cr are calculated by comparing the integral area. At the same time, the pseudochromatic PWI images were obtained on Mr postprocessing workstation, and the rCBV values around the bilateral ventricles of rats were measured after perfusion. Results no abnormal MRI was found in the brain of all the examined rats. 2 the NAA/Cr ratio of the rats in each irradiation group decreased with time, compared with that of the control group. The ratio of Cho/Cr increased firstly and reached the peak at 3 months after irradiation. The Cho/Cr of each group was significantly lower than that of control group within 6 months. The rCBV of rats in the group 1 month after P0.05.3 irradiation had no statistical significance compared with that of the control group. The rCBV value of each group was significantly lower than that of the control group one month after irradiation, and the difference was statistically significant (P 0.05). Conclusion 1 Mrs can detect the abnormal metabolism of brain tissue before the morphological changes of radiation brain injury. 2PWI can detect the abnormal microcirculation of brain tissue before the morphological changes of radiation brain injury.
