運(yùn)動(dòng)訓(xùn)練和表沒(méi)食子兒茶素沒(méi)食子酸酯對(duì)2型糖尿病大鼠腎臟TLR4炎性因子表達(dá)的作用研究

【摘要】:目的探討運(yùn)動(dòng)訓(xùn)練、表沒(méi)食子兒茶素沒(méi)食子酸酯(EGCG)單獨(dú)干預(yù)及聯(lián)合應(yīng)用對(duì)2型糖尿病大鼠腎臟血管通透性和腎TLR4、NF-κBp65、IL-1β蛋白表達(dá)量的影響。方法通過(guò)高脂喂養(yǎng)加腹腔注射小劑量鏈脲佐菌素(STZ)構(gòu)建2型糖尿病SD大鼠模型。將造模成功的56只大鼠隨機(jī)分為糖尿病對(duì)照組(D)、糖尿病給藥組(DY)、糖尿病運(yùn)動(dòng)組(DE)、糖尿病運(yùn)動(dòng)給藥組(DEY),每組14只,另設(shè)正常對(duì)照組(C)12只。運(yùn)動(dòng)組大鼠進(jìn)行12周不負(fù)重游泳訓(xùn)練,每周訓(xùn)練5 d。EGCG給藥組大鼠每周灌胃7 d。實(shí)驗(yàn)結(jié)束后檢測(cè)大鼠FBG、尿蛋白、UAER、TLR4、NF-κBp65、IL-1β蛋白和腎臟血管通透性變化,并對(duì)實(shí)驗(yàn)結(jié)果進(jìn)行統(tǒng)計(jì)分析。結(jié)果與正常對(duì)照組(C)相比,糖尿病對(duì)照組(D)的FBG、尿蛋白、UAER、TLR4、NF-κBp65、IL-1β蛋白和腎臟血管通透性均顯著升高(P0.01);糖尿病給藥組(DY)、糖尿病運(yùn)動(dòng)組(DE)、糖尿病運(yùn)動(dòng)給藥組(DEY)的FBG、尿蛋白、UAER、TLR4、NF-κBp65、IL-1β蛋白和腎臟血管通透性較糖尿病對(duì)照組(D)均顯著降低(P0.05或P0.01);糖尿病運(yùn)動(dòng)給藥組(DEY)FBG、UAER、TLR4、NF-κBp65、IL-1β蛋白表達(dá)量和腎血管通透性均顯著低于糖尿病給藥組(DY)(P0.05或P0.01),糖尿病運(yùn)動(dòng)給藥組(DEY)腎臟指數(shù)、尿蛋白、UAER、TLR4、NF-κBp65、IL-1β蛋白表達(dá)量和腎血管通透性均顯著低于糖尿病運(yùn)動(dòng)組(DE)(P0.05)。結(jié)論 2型糖尿病大鼠存在腎組織炎性因子表達(dá)增多,運(yùn)動(dòng)訓(xùn)練和EGCG可改善2型糖尿病大鼠腎臟炎性損傷,抑制TLR4信號(hào)通路。
[Abstract]:Objective to investigate the effects of exercise training, epigallocatechin gallate (EGCGG) alone and combined therapy on renal vascular permeability and renal TLR4NF- 魏 Bp65 / IL-1 尾 protein expression in type 2 diabetic rats. Methods SD rat model of type 2 diabetes was established by high fat feeding and intraperitoneal injection of streptozotocin (STZ). Fifty-six rats were randomly divided into three groups: control group (n = 14), control group (n = 12), diabetic exercise group (n = 14), diabetic exercise group (n = 14) and diabetic exercise group (n = 14). The rats in the exercise group were trained for 12 weeks without weight bearing swimming, and the rats in the control group were given 5 d.EGCG weekly intragastric perfusion for 7 days. At the end of the experiment, the changes of FBG, urinary protein TLR4, NF- 魏 Bp65, IL-1 尾 and renal vascular permeability in rats were detected and the results were statistically analyzed. Results compared with the normal control group, FBG, urinary protein TLR4, NF- 魏 Bp65, IL-1 尾 and renal vascular permeability in diabetic control group were significantly higher than those in diabetic group, diabetic exercise group and diabetic exercise group. Urinary protein UAERTLR4NF-Bp65 尾 and renal vascular permeability were higher than those in diabetic group. The expression of IL-1 尾 protein and renal vascular permeability in diabetic exercise group were significantly lower than those in diabetic group (P 0.05 or P 0.01), and the renal index in diabetic exercise group was significantly lower than that in diabetic exercise group (P 0.05 or P 0.01), and the expression of IL-1 尾 protein in TLR4 and renal vascular permeability was significantly lower in diabetic exercise group than that in diabetic exercise group, and the expression of IL-1 尾 and renal vascular permeability were significantly lower in the control group than in the diabetic exercise group, and the renal vascular permeability was significantly lower in the diabetic exercise group than that in the diabetic exercise group. The expression of NF- 魏 Bp65- 尾 and renal vascular permeability in UAR4 were significantly lower than those in diabetic exercise group (P 0.05). Conclusion there is an increase in the expression of renal inflammatory factors in type 2 diabetic rats. Exercise training and EGCG can improve the renal inflammatory injury and inhibit the TLR4 signal pathway in type 2 diabetic rats.
