自擬養(yǎng)陰生津方對(duì)輻射損傷小鼠HPRT基因防護(hù)作用研究

【摘要】:目的: 探討自擬養(yǎng)陰生津方對(duì)輻射損傷小鼠hprt基因防護(hù)作用,豐富中醫(yī)藥抗輻射理論,為中醫(yī)藥方劑預(yù)防放射輻射和降低放射輻射所導(dǎo)致的損害提供可檢測(cè)的實(shí)驗(yàn)方法和理論指導(dǎo)。 方法: 動(dòng)物分組:取雄性健康昆明種小鼠200只隨機(jī)分為五組,每組40只。A.生理鹽水組、B.照射1組、C.照射2組、D.中藥1組、E.中藥2組;照射:一次性給予B組、D組8Gy劑量的X射線。多次給予C組、E組總劑量為8Gy的X射線,1Gy/d連續(xù)照射8天。給藥:給D.組、E.組小鼠用自擬養(yǎng)陰生津方灌胃,1次/d,0.5ml/次/只.其余各組小鼠用生理鹽水以同樣方法灌胃,連續(xù)10天,然后照射。照射期及其后繼續(xù)給藥,直至該組實(shí)驗(yàn)結(jié)束。檢測(cè)及觀察:照射結(jié)束后第5天,每組取10只小鼠處死,檢測(cè)各組小鼠hprt基因突變率和突變譜。觀察每組30只小鼠的30天存活率及平均存活天數(shù)。 結(jié)果: 1.小鼠的存活率及平均存活天數(shù)。各組小鼠30天存活率分別:A.生理鹽水組100%,B.照射1組0.00%,C.照射2組13.3%,D.中藥1組16.7%, E.中藥2組26.7%。各組小鼠平均存活天數(shù)分別為:A.生理鹽水組30.0±0.0天,B.照射1組8.3±1.3天,C.照射2組13.1±4.2天,D.中藥1組12.1±3.3天, E.中藥2組16.4±1.7天。照射1組、照射2組小鼠30天存活率及平均存活天數(shù)與生理鹽水組比較均明顯降低,差異有顯著性(P0.01);給予小鼠自擬養(yǎng)陰生津方后,中藥1組存活率及平均存活天數(shù)與照射1組比較有明顯提高,差異有顯著性(P0.05);中藥2組存活率及平均存活天數(shù)與照射2組比較有明顯提高,差異有顯著性(P0.05)。 2.各組小鼠外周血淋巴細(xì)胞hprt基因的突變率分別為:A.生理鹽水組0.35±0.02‰,B.照射1組0.80±0.07‰,C.照射2組0.98±0.02‰,D.中藥1組0.72±0.02‰,E.中藥2組0.85±0.01‰。照射1組、2組小鼠hprt基因的突變率與生理鹽水組比較均明顯升高,差異有顯著性(P0.01);給予小鼠自擬養(yǎng)陰生津方后,中藥1組小鼠hprt基因的突變率與照射1組比較有明顯降低,差異有顯著性(P0.05);中藥2組小鼠hprt基因的突變率與照射2組比較有明顯降低,差異有顯著性(P0.05)。 3.各組小鼠外周血淋巴細(xì)胞hprt基因的突變譜缺失突變比例分別為:A.生理鹽水組25%,B.照射1組75%, C.照射2組87.5%,, D.中藥1組50%,E.中藥2組62.5%。照射1組、2組小鼠hprt基因的突變譜缺失突變比例與生理鹽水組比較均明顯升高,差異有顯著性(P0.01);給予小鼠自擬養(yǎng)陰生津方后,中藥1組小鼠hprt基因的突變譜缺失突變比例與照射1組比較有明顯降低,差異有顯著性(P0.05);中藥2組小鼠hprt基因的突變譜缺失突變比例與照射2組比較有明顯降低,差異有顯著性(P0.05)。 結(jié)論: 1.自擬養(yǎng)陰生津方能提高輻射小鼠的30天存活率及平均存活天數(shù)。 2.自擬養(yǎng)陰生津方能降低輻射小鼠hprt基因突變率。 3.自擬養(yǎng)陰生津方能降低輻射小鼠hprt基因突變譜缺失突變比例。
[Abstract]:Purpose :
This paper discusses the protective effect of self - prepared yin - generating body on hprf gene in radiation - injured mice , enriches the theory of anti - radiation of traditional Chinese medicine , and provides the experimental method and theoretical guidance for the prevention of radiation radiation and the reduction of radiation induced by Chinese medicinal prescription .
Method :
Animal grouping : 200 male healthy Kunming mice were randomly divided into five groups : 40 . A . physiological saline group , B . irradiation group 1 , C . irradiation group 2 , D . Chinese medicine 1 group , E . traditional Chinese medicine 2 group ;
Two groups of mice were treated with normal saline for 8 days . The total dose of group E was 8Gy , and the dose of 1Gy / d was given for 8 days . The results were as follows : 5 days after the end of irradiation , 10 mice were sacrificed and the mutation rate and mutation spectrum of the mice were detected . The survival rate and the average survival days of 30 days in each group were observed .
Results :
1 . The survival rate and the average survival days of mice in each group were 100 % and 100 % in group A , 0.00 % in group B , 13.3 % in group C , 16.7 % in group B , and 26.7 % in group E . The average survival days of mice in each group were as follows : A . Saline group 30.0 鹵 0 . 0 days , D . Chinese medicine 1 group 12.1 鹵 3.3 days , E . Chinese medicine group 2 group 16.4 鹵 1.7 days . The survival rate and average survival days of irradiated group 2 mice were significantly lower than those in physiological saline group , and the difference was significant ( P0.01 ) .
The survival rate and the average survival time of the traditional Chinese medicine group were significantly increased compared with those in the irradiated group ( P0.05 ) .
The survival rate and average survival time of the traditional Chinese medicine group were significantly improved compared with those in group 2 ( P0.05 ) .
2 . The mutation rate of hpr1 gene in peripheral blood lymphocytes of each group was 0 . 35 鹵 0 . 02 鈥
