耐力訓練對帕金森模型小鼠中腦線粒體自噬相關基因表達的影響

【摘要】:目的:探討耐力訓練對帕金森病(PD)模型小鼠中腦線粒體自噬的影響,揭示運動預防帕金森病的分子生物學機制。方法:10周齡雄性C57BL/6小鼠32只,隨機分為安靜組(C),運動組(E),帕金森模型安靜組(P)及帕金森模型運動組(PE),每組8只。E組及PE組小鼠進行為期8周跑臺耐力訓練,跑臺速度15m/min,坡度為0%,每天運動持續(xù)50min,每周6次,周日休息。訓練結束后,P組和PE組小鼠腹腔注射MPTP(30mg/kg),C組和E組小鼠注射等量生理鹽水,持續(xù)3d。注射結束1周后,處死各組小鼠,取雙側中腦及紋狀體,采用實時PCR及Western blot技術檢測線粒體自噬相關基因及TH蛋白表達,HPLC技術檢測中腦及紋狀體組織內DA含量。結果:與C組比,E組小鼠中腦TH蛋白上調(P0.05),中腦及紋狀體DA含量均增加(P0.05),P組和PE組小鼠TH蛋白及DA含量均顯著下降(P0.01),而P組較PE組相應指標下降更明顯(P0.05)。與C組比,E組小鼠中腦PINK、PARK2 m RNA及PINK1蛋白表達上調(P0.05),LC3Ⅱ/Ⅰ比值增加,P組小鼠PINK1m RNA表達下調(P0.05),PE組小鼠PINK1 m RNA、Parkin蛋白含量及LC3Ⅱ/Ⅰ比值較P組增加。結論:為期3d的MPTP處理可導致小鼠中腦及紋狀體TH及DA含量下降,誘導帕金森病發(fā)生,耐力訓練可緩解這一現(xiàn)象;MPTP誘導的帕金森小鼠模型中腦線粒體自噬水平下降,而耐力訓練可作用于線粒體自噬相關基因的表達,抑制MPTP的神經(jīng)毒性作用。
[Abstract]:Aim: to investigate the effects of endurance training on mitochondrial autophagy in midbrain of PD mice and to reveal the molecular biological mechanism of exercise in preventing Parkinson's disease. Methods Thirty-two male 10 week-old male C57BL/6 mice were randomly divided into three groups: control group, exercise group, Parkinson model group (P) and Parkinson's model exercise group (P). Each group of 8 mice in .E group and PE group were trained for 8 weeks. Running at a speed of 15 m / min, with a slope of 0, running 50 minutes a day, six times a week, and resting on Sundays. After training, mice in P group and PE group were intraperitoneally injected with 30 mg / kg MPTPN in group C and E, respectively. The same amount of normal saline was injected into mice of group E for 3 days. One week after the injection, the mice were killed and the contents of DA in the midbrain and striatum were detected by real-time PCR and Western blot. The expression of th protein and mitochondrial autophagy related gene were detected by HPLC. Results: compared with group C, th protein in mesencephalon of mice in E group was up-regulated, and DA content in midbrain and striatum were increased. Th protein and DA content in P0.05 group and PE group were significantly decreased compared with that in group C, while in group P, the decrease of corresponding indexes in group P was more obvious than that in group PE. Compared with group C, the expression of PINKT PARK2 m RNA and PINK1 protein in the midbrain of E group was increased compared with that in P group. The increase of PINK1m RNA expression in P group was higher than that in P group. Compared with that in P group, the expression of PINK1m RNA in P group decreased PINK1 m RNA-parkin protein content and the ratio of LC3 鈪,
