紫甘薯花青素對過度訓(xùn)練大鼠心肌氧化損傷干預(yù)的研究

【摘要】:研究目的: 研究表明,自由基是過度訓(xùn)練對大鼠心肌氧化損傷的最直接物質(zhì),服用抗氧化性物質(zhì)可以減少自由基的生成,降低心肌受到的氧化損傷。本研究以抗氧化物(紫甘薯花青素)對過度訓(xùn)練大鼠心肌氧化損傷的干預(yù)為切入點(diǎn),分析抗氧化物(紫甘薯花青素)對過度訓(xùn)練大鼠心肌氧化損傷的影響,以尋求新的抗心肌氧化損傷的物質(zhì),豐富有關(guān)抗心肌氧化損傷的物質(zhì)來源。本研究旨在探討紫甘薯花青素的抗氧化作用對過度訓(xùn)練大鼠心肌氧化損傷的干預(yù)效果,為進(jìn)一步研究紫甘薯花青素等抗氧化物對過度訓(xùn)練引起的心肌損傷的干預(yù)提供一個(gè)可靠的依據(jù)。 研究方法: 本研究采用動(dòng)物實(shí)驗(yàn)法,正式實(shí)驗(yàn)之前先進(jìn)行預(yù)實(shí)驗(yàn),以健康雄性SD大鼠為實(shí)驗(yàn)對象。預(yù)實(shí)驗(yàn)選取較成熟的過度訓(xùn)練方案,進(jìn)行六周的造模,然后取大鼠腹主動(dòng)脈血液制備血清,測量判斷過度訓(xùn)練的主指標(biāo):血睪酮(T)和皮質(zhì)醇(C),通過分析指標(biāo),得出預(yù)實(shí)驗(yàn)中所用的訓(xùn)練方案能夠成功的復(fù)制過度訓(xùn)練模型,該方案可以應(yīng)用于正式實(shí)驗(yàn)當(dāng)中。正式實(shí)驗(yàn)選取健康雄性SD大鼠84只,隨機(jī)分為4大組,分別為空白對照組(C組)、一般訓(xùn)練組(NS組)、過度訓(xùn)練組(OT組)和過度訓(xùn)練花青素組(OTH組),其中所有運(yùn)動(dòng)組大鼠分兩種處死狀態(tài),即安靜狀態(tài)下處死(后加1表示)和即刻狀態(tài)下處死(后加2表示)。運(yùn)動(dòng)組大鼠周日至周五運(yùn)動(dòng),周六休息,共進(jìn)行6周,然后取大鼠腹主動(dòng)脈血液制備血清并摘取心臟,零下80℃保存。實(shí)驗(yàn)結(jié)束后,制作心肌組織切片并進(jìn)行HE染色病理學(xué)觀察,測量分析血清指標(biāo)心肌肌鈣蛋白I(cTnI)、肌酸激酶同工酶(CK-MB)、乳酸脫氫酶(LDH)、肌酸激酶(CK)、血睪酮(T)和皮質(zhì)醇(C),大鼠心肌線粒體指標(biāo)丙二醛(MDA)、超氧化物歧化酶(SOD)、線粒體內(nèi)Ca2+濃度(Ca2+),對檢測結(jié)果進(jìn)行相關(guān)統(tǒng)計(jì)學(xué)分析。 研究結(jié)果: 1NS組超氧化物歧化酶、丙二醛、Ca2+濃度、血睪酮、皮質(zhì)醇、乳酸脫氫酶、肌酸激酶、肌酸激酶同工酶、心肌肌鈣蛋白I與C組對比均沒有差異性;NS組與C組相比,肌纖維增粗,排列整齊有序。 2OT組超氧化物歧化酶、血睪酮與C組和NS組相比均顯著降低,丙二醛、Ca2+濃度、皮質(zhì)醇、乳酸脫氫酶、肌酸激酶、肌酸激酶同工酶、心肌肌鈣蛋白I的水平均顯著高于C組和NS組;組織切片表明OT組與C組和NS組相比,心肌過度肥大,肌纖維排列疏松錯(cuò)亂,有病態(tài)現(xiàn)象。 3OTH組超氧化物歧化酶、血睪酮水平顯著低于C組和NS組;丙二醛、皮質(zhì)醇、Ca2+濃度、乳酸脫氫酶、肌酸激酶、肌酸激酶同工酶、心肌肌鈣蛋白I的水平均顯著高于C組和NS組;OTH組超氧化物歧化酶、血睪酮水平高于OT組,但不顯著;丙二醛、Ca2+濃度、皮質(zhì)醇、乳酸脫氫酶、肌酸激酶、肌酸激酶同工酶、心肌肌鈣蛋白I的水平低于OT組,其中心肌肌鈣蛋白I、Ca2+濃度差異非常顯著,其他指標(biāo)顯著。OTH組的組織切片也表現(xiàn)出了一定的病態(tài)現(xiàn)象,但是要輕于OT組。 研究結(jié)論: 1適宜的運(yùn)動(dòng)訓(xùn)練能一定程度的提高心肌的抗氧化能力,能使心肌肌纖維增粗,排列更致密有序,引導(dǎo)心肌向著健康的方向發(fā)展,對心肌具有一定的保護(hù)作用。 2過度訓(xùn)練破壞了大鼠心肌內(nèi)自由基的防御系統(tǒng),心肌細(xì)胞結(jié)構(gòu)被破壞,肌纖維過度肥大,排列錯(cuò)亂無規(guī)則,心肌組織受到一定程度的氧化損傷,有病態(tài)現(xiàn)象。 3紫甘薯花青素的干預(yù)能一定程度的提高過度訓(xùn)練大鼠的抗氧化能力,保護(hù)心肌細(xì)胞的結(jié)構(gòu),減輕肌纖維的排列錯(cuò)亂現(xiàn)象,病態(tài)現(xiàn)象減輕,降低了過度訓(xùn)練所引起的心肌氧化損傷程度。 4經(jīng)過24小時(shí)的休息,一般訓(xùn)練大鼠心肌的輕微損傷能得到有效的修復(fù);過度訓(xùn)練運(yùn)動(dòng)大鼠也有所修復(fù),但是修復(fù)程度較。蛔细适砘ㄇ嗨氐母深A(yù)可以在一定程度上加快過度訓(xùn)練大鼠心肌氧化損傷的修復(fù)速度。
[Abstract]:The purpose of the study is:
Studies have shown that free radicals are the most direct substances of oxidative damage to rats' Myocardium by overtraining. Taking antioxidant substances can reduce the formation of free radicals and reduce oxidative damage to the myocardium. The effect of sweet potato anthocyanins on oxidative damage of myocardium in overtrained rats, in order to seek new substances against myocardial oxidative damage, and to enrich the material sources of anti myocardial oxidative damage. The aim of this study was to explore the effect of antioxidant effects of purple sweet potato anthocyanins on myocardial oxygen damage in overtrained rats and to further study purple sweet potato Anthocyanins and other antioxidants provide a reliable basis for the intervention of myocardial injury induced by overtraining.
Research methods:
In this study, the animal experiment method was used. Before the formal experiment, a healthy male SD rat was taken as the experimental object. A more mature overtraining program was selected to make a model for six weeks. Then the blood serum was prepared from the abdominal aorta of rats. The main index of overtraining was measured: blood testosterone (T) and cortisol (C). It can be used in the formal experiment. 84 healthy male SD rats were randomly divided into 4 groups: blank control group (group C), general training group (group NS), overtraining group (OT group) and overtraining anthocyanin group. (group OTH), all of the rats in the exercise group were killed in two kinds of death States, that is, to be executed in a quiet state (after adding 1) and to be executed under immediate condition. The rats in the exercise group were exercise on Sunday to Friday, and the Saturday rest, for a total of 6 weeks. Then the rat abdominal aorta was taken to prepare the serum and extract the heart and save at minus 80. After the experiment, the experiment was made. Myocardial tissue sections and HE staining pathological observation were performed to measure and analyze serum cardiac troponin I (cTnI), creatine kinase isoenzyme (CK-MB), lactate dehydrogenase (LDH), creatine kinase (CK), serum testosterone (T) and cortisol (C), myocardial mitochondrial index of malondialdehyde (MDA), superoxide dismutase (SOD), and Ca2+ concentration in mitochondria (Ca2+). Statistical analysis was carried out on the results of the test.
The results of the study:
1NS group superoxide dismutase, malondialdehyde, Ca2+ concentration, blood testosterone, cortisol, lactate dehydrogenase, creatine kinase, creatine kinase isoenzyme, cardiac troponin I and C group were no difference, NS group and C group, muscle fiber thickened, arranged orderly and orderly.
The level of MDA, Ca2+, cortisol, creatine kinase, creatine kinase isoenzyme and cardiac troponin I were significantly higher in group 2OT than in group C and NS. The tissue section showed that OT group was hyperhypertrophy of myocardium and loosely arranged muscle fiber in group OT and C group and NS group. A disorder, a pathological phenomenon.
3OTH group superoxide dismutase, blood testosterone level was significantly lower than group C and NS, malondialdehyde, cortisol, Ca2+ concentration, lactate dehydrogenase, creatine kinase, creatine kinase isoenzyme, myocardial troponin I level were significantly higher than the C and NS groups; OTH group superoxide dismutase, blood testosterone level was higher than the OT group, but not significant; malondialdehyde, Ca2+ concentration Degree, cortisol, lactate dehydrogenase, creatine kinase, creatine kinase isoenzyme, cardiac troponin I level were lower than that of group OT, and the difference of cardiac troponin I and Ca2+ concentration was very significant. Other indicators also showed a certain pathological phenomenon in group.OTH, but it should be lighter than group OT.
The conclusions are as follows:
1 suitable exercise training can improve the antioxidant capacity of the myocardium to a certain extent, make the muscle fiber thickening, arrange more compact and orderly, guide the heart to the direction of health and have a certain protective effect on the myocardium.
2 over training destroys the free radical defense system in the rat myocardium, the structure of the myocardial cells is destroyed, the muscle fiber hypertrophy, the disorder of the arrangement is irregular, the myocardial tissue is damaged by some degree of oxidative damage, and there is a pathological phenomenon.
The intervention of 3 purple sweet potato anthocyanins can improve the antioxidant capacity of overtraining rats to a certain extent, protect the structure of cardiac myocytes, reduce the disorder of muscle fiber arrangement, reduce the pathological phenomenon, and reduce the degree of myocardial oxidative damage caused by overtraining.
4 after 24 hours of rest, the mild injury of the rat's myocardium can be effectively repaired, and the overtraining exercise rats are also repaired, but the repair degree is small, and the intervention of Purple Sweet Potato Anthocyanin can accelerate the repair speed of myocardial oxidative damage in rats to a certain extent.
