茄尼醇對(duì)大鼠運(yùn)動(dòng)疲勞恢復(fù)期心肌自由基代謝和血清相關(guān)指標(biāo)的影響

【摘要】:大強(qiáng)度運(yùn)動(dòng)過程中,機(jī)體內(nèi)自由基生成量增加,自身的抗氧化系統(tǒng)難以清除過剩的自由基,是運(yùn)動(dòng)損傷和疲勞發(fā)生的重要原因。外源性抗氧化劑的補(bǔ)充可延緩或逆轉(zhuǎn)由于氧化應(yīng)激引起的各種慢性病理過程及運(yùn)動(dòng)性疲勞的發(fā)生和發(fā)展,提高運(yùn)動(dòng)尤其是有氧運(yùn)動(dòng)能力。茄尼醇是一種不飽和的聚異戊二烯醇,屬四倍半萜醇。茄尼醇本身具有9個(gè)異戊二烯基團(tuán)和多個(gè)非共軛雙鍵,因此具有非常強(qiáng)烈的吸收自由基的性能。茄尼醇可以作為異戊二烯單元用于合成具有生物活性的泛醌類物質(zhì),如輔酶Q_(10)和維生素K_2類。而輔酶Q_(10)是一種代謝激活劑,可以改變細(xì)胞和組織的缺氧狀態(tài),同時(shí),輔酶Q_(10)還是細(xì)胞自身產(chǎn)生的天然抗氧化劑,能抑制線粒體的過氧化,保護(hù)生物膜結(jié)構(gòu)的完整性。 實(shí)驗(yàn)?zāi)康模涸谶\(yùn)動(dòng)訓(xùn)練中,運(yùn)動(dòng)性疲勞的產(chǎn)生、積累和消除是一個(gè)連續(xù)過程。本文通過大鼠運(yùn)動(dòng)實(shí)驗(yàn),研究在運(yùn)動(dòng)與恢復(fù)期間疲勞的產(chǎn)生、積累和清除這一漸進(jìn)的、連續(xù)的過程中,補(bǔ)充茄尼醇對(duì)遞增運(yùn)動(dòng)負(fù)荷訓(xùn)練至力竭的運(yùn)動(dòng)性疲勞大鼠心肌和血清相關(guān)指標(biāo)影響及其規(guī)律,旨在為探討補(bǔ)充茄尼醇對(duì)延緩運(yùn)動(dòng)性疲勞的產(chǎn)生和加速疲勞的消除以及對(duì)大鼠心肌細(xì)胞的保護(hù)作用提供實(shí)驗(yàn)依據(jù)。 實(shí)驗(yàn)方法:實(shí)驗(yàn)以健康雄性SD大鼠為研究對(duì)象,以遞增負(fù)荷跑臺(tái)訓(xùn)練方式建立運(yùn)動(dòng)性疲勞模型。將跑臺(tái)適應(yīng)訓(xùn)練、篩選后大鼠隨機(jī)分為:安靜組(UT);運(yùn)動(dòng)即刻組(TC1)、恢復(fù)6h組(TC2)和恢復(fù)24h組(TC3);運(yùn)動(dòng)小劑量即刻組(TST1)、恢復(fù)6h組(TST2)和恢復(fù)24h組(TST3);運(yùn)動(dòng)大劑量即刻組(TLT1)、恢復(fù)6h組(TLT2)和恢復(fù)24h組(TLT3)。劑量組每天上午以按照藥劑量3mg/100g(小劑量)、6mg/100g(大劑量)給予灌胃,下午按照運(yùn)動(dòng)疲勞方案進(jìn)行訓(xùn)練,除安靜組外,其余各組按遞增負(fù)荷訓(xùn)練2周,中間不休息,2周訓(xùn)練后第1天,大鼠除安靜組外,所有其他組最后一次全部以30m/min的速度一次性力竭。大鼠力竭后,按照不同恢復(fù)時(shí)間,摘除眼球采血,然后斷髓處死,同時(shí)迅速取出心臟,分別檢測大鼠心肌組織SOD、GSH-Px、MDA、CAT、SDH、LDH和血清的BUN和CK。 實(shí)驗(yàn)結(jié)果: (1)運(yùn)動(dòng)力竭即刻,TST心肌組織SOD、GSH-PX、CAT、LDH、SDH的活性和SOD/MDA比值顯著性(P0.01)高于TC,TLT也有所升高,與TC和TST相比,無顯著性差異;TST心肌組織MDA活性顯著性(P0.01)低于TC和TLT,TST血清CK活性最低,與TC和TLT無顯著性差異,TST血清BUN顯著性(P0.05)低于TC,與TLT無顯著性差異。 (2)運(yùn)動(dòng)力竭恢復(fù)6h, TST心肌組織GSH-PX、LDH、SDH的活性和SOD/MDA比值顯著性(P0.01)高于TC,且顯著性(P0.05)高于TLT;TST血清CK、BUN活性顯著性(P0.05)低于TC,且TST血清BUN顯著性(P0.05)低于TLT。 (3)運(yùn)動(dòng)力竭恢復(fù)24h, TST心肌組織GSH-PX、CAT、LDH、SDH的活性和SOD/MDA比值顯著性(P0.01)高于TC,其中,TST心肌組織CAT、LDH、的活性和SOD/MDA比值顯著性(P0.01)高于TLT;TST血清CK、BUN活性顯著性(P0.01)低于TC,且TST血清BUN顯著性(P0.05)低于TLT。 實(shí)驗(yàn)結(jié)論: (1)外源性補(bǔ)充外源性茄尼醇可不同程度提高運(yùn)動(dòng)大鼠恢復(fù)期心肌SOD/MDA比值GSH-PX和CAT活性。說明茄尼醇具有良好的抗脂質(zhì)過氧化作用,可以降低由大量自由基引起的心肌缺血再灌注損傷程度和心肌細(xì)胞凋亡率,保護(hù)心肌細(xì)胞膜結(jié)構(gòu)和功能完整性,促進(jìn)運(yùn)動(dòng)疲勞的恢復(fù)。 (2)茄尼醇可不同程度提高運(yùn)動(dòng)大鼠恢復(fù)期心肌LDH、SDH的活性,降低血清CK活性和尿素氮的含量,,從而緩解疲勞產(chǎn)生、增強(qiáng)運(yùn)動(dòng)能力;有利于有氧代謝過程,促進(jìn)運(yùn)動(dòng)時(shí)心肌能量的供給和疲勞恢復(fù)。 (3)小劑量對(duì)大鼠運(yùn)動(dòng)疲勞恢復(fù)的影響優(yōu)于大劑量。
[Abstract]:Coenzyme Q _ ( 10 ) is a kind of unsaturated polyisoprenoid , which can delay or reverse the occurrence and development of various chronic diseases , such as coenzyme Q _ ( 10 ) and vitamin K _ 2 , which are caused by oxidative stress .
The aim of this study is to study the effects of solanesol on cardiac and serum - related indexes of exercise - induced fatigue in rats during exercise and recovery , and to provide experimental basis for the study of the effects of solanesol on the delayed exercise fatigue and the elimination of accelerated fatigue .
The experimental methods were as follows : the healthy male SD rats were used as the research objects , and the sports fatigue model was established by increasing the training mode of the load running platform . The treadmill was adapted to the training , and the rats after screening were randomly divided into : quiet group ( UT ) ;
exercise immediate group ( tcl ) , recover 6h group ( tc2 ) and recover 24h group ( tc3 ) ;
Immediate group ( TST1 ) , 6h group ( TST2 ) and 24h group were recovered ( Tst3 ) .
The rats were divided into three groups according to the daily dose of 3 mg / 100g ( small dose ) , 6 mg / 100g ( high dose ) and 6 mg / 100g ( high dose ) . The rats were divided into three groups according to the daily dose of 3 mg / 100g ( small dose ) and 6 mg / 100g ( high dose ) .
Experimental results :
( 1 ) Immediately after exercise , the activity of SOD , GSH - PX , CAT , LDH , SDH and SOD / MDA in myocardium were significantly higher than those of TC and TLT ( P0.01 ) .
There was no significant difference in serum CK activity between TLT and TLT ( P0.01 ) . There was no significant difference with TC and TLT . There was no significant difference in serum BUN ( P0.05 ) between TLT and TLT .
( 2 ) The activity of GSH - PX , LDH , SDH and the ratio of SOD / MDA were significantly higher than that of TC ( P0.01 ) .
There was significant difference ( P0.05 ) between serum CK and BUN ( P0.05 ) , and the serum BUN was lower than that of TLT ( P0.05 ) .
( 3 ) The activity of GSH - PX , CAT , LDH , SDH and the ratio of SOD / MDA ( P0.01 ) were higher than that of TC , and the activities of CAT , LDH , LDH , LDH , SDH and SOD / MDA were significantly higher than that of TLT ( P0.01 ) .
There was significant difference ( P0.01 ) between serum CK and BUN ( P0.01 ) , and the serum BUN was lower than that of TLT ( P0.05 ) .
Experimental Conclusion :
( 1 ) The activities of SOD / MDA and GSH - PX and CAT in the myocardium of rats with different degrees could be increased in different degrees . The results showed that solanesol had a good anti - lipid peroxidation effect , and could reduce the degree of myocardial ischemia reperfusion injury induced by a large number of free radicals and the apoptosis rate of myocardial cells , protect the membrane structure and functional integrity of myocardial cells , and promote the recovery of exercise fatigue .
( 2 ) solanesol can improve the activity of LDH and SDH in the recovery period of exercise rats in different degrees , reduce the serum CK activity and the content of urea nitrogen , thereby relieving fatigue and enhancing athletic ability ;
is beneficial to the aerobic metabolism process and promotes the supply and fatigue recovery of myocardial energy during exercise .
( 3 ) The effect of small dose on the recovery of exercise fatigue in rats was superior to that of large dose .
