維生素E聯(lián)合紅景天對低氧復(fù)合運動小鼠骨骼肌保護作用的研究

【摘要】:目的探討維生素E聯(lián)合紅景天對低氧復(fù)合運動小鼠骨骼肌細(xì)胞損傷的保護性作用及其機制。方法將40只ICR小鼠按照隨機數(shù)字表法分為對照組和觀察組,每組均為20只。對照組小鼠給予灌胃維生素E(40mg·kg~(-1)·d~(-1)),觀察組小鼠給予灌胃維生素E(40mg·kg~(-1)·d~(-1))+紅景天提取液(20g·kg~(-1)·d~(-1)),兩組小鼠連續(xù)給藥15d后進行為期4周的低氧(氧濃度11.3%)復(fù)合運動(5℃,15m/min,60min/d)。最后一次運動后處死小鼠檢測骨骼肌血清甘肽過氧化物酶(GSH-PX)、超氧化物歧化酶(SOD)、活性氧類物質(zhì)(ROS)、丙二醛(MDA);逆轉(zhuǎn)錄PCR(RT-PCR)法檢測骨骼肌B淋巴細(xì)胞瘤-2(Bcl-2)和Bcl-2相關(guān)X蛋白(Bax)基因mRNA水平的表達,進行氧復(fù)合運動過程中(運動前、1、2、4周)監(jiān)測外周血清肌酸激酶(CK)、乳酸脫氫酶(LDH)。結(jié)果運動后各時間點觀察組CK與LDH活性均低于對照組(P0.05)。觀察組小鼠骨骼肌組織的GSH-PX、SOD活性明顯高于對照組(P0.05),而ROS、MDA低于對照組(P0.05);觀察組小鼠骨骼肌組織Bax基因mRNA水平的表達與Bax/Bcl-2比值均明顯低于對照組(P0.05),而Bcl-2基因mRNA水平的表達明顯高于對照組(P0.05)。結(jié)論紅景天聯(lián)合維生素E能通過糾正低氧運動下的氧化與抗氧化失衡而抑制凋亡基因的表達。
[Abstract]:Objective to investigate the protective effect and mechanism of vitamin E combined with Rhodiola on skeletal muscle cell injury in hypoxic combined exercise mice.Methods 40 ICR mice were randomly divided into control group and observation group with 20 in each group.The expression of gene mRNA,During the oxygenation exercise (1 ~ 2 weeks before exercise), the levels of creatine kinase (CK) and lactate dehydrogenase (LDH) in peripheral blood were monitored.Results the activities of CK and LDH in the observation group were lower than those in the control group at each time point after exercise.The activity of GSH-PXO SOD in skeletal muscle of the observation group was significantly higher than that of the control group (P 0.05), while the rosid MDA was lower than that of the control group (P 0.05). The expression of Bax gene mRNA and the ratio of Bax/Bcl-2 to Bax/Bcl-2 in the skeletal muscle of the observation group were significantly lower than those in the control group (P 0.05), while the mRNA level of Bcl-2 gene in the observation group was significantly lower than that in the control group.The expression of P0.05 in the control group was significantly higher than that in the control group.Conclusion Rhodiola combined with vitamin E can inhibit the expression of apoptotic genes by correcting the imbalance of oxidation and antioxidation in hypoxic exercise.
