益美膚治療SD大鼠急性β射線皮膚Ⅱ度損傷創(chuàng)面的實(shí)驗(yàn)研究

【摘要】:目的:通過(guò)比較創(chuàng)面大體變化、組織學(xué)變化及表皮干細(xì)胞標(biāo)記物K19及整合素β1陽(yáng)性細(xì)胞表達(dá)方面的變化,探討益美膚對(duì)SD大鼠皮膚急性β射線Ⅱ度皮膚損傷創(chuàng)面的修復(fù)的效果。方法:2-3月齡清潔級(jí)SD.大鼠102只,體重190±10g,單籠喂養(yǎng)5天以適應(yīng)環(huán)境。實(shí)驗(yàn)動(dòng)物創(chuàng)面制備時(shí)使用SIMENS直線加速器,參數(shù)設(shè)置6Me V高能電子束,單次照射大鼠臀部裸露皮膚11分鐘52秒,制作SD大鼠急性β射線Ⅱ度皮膚損傷創(chuàng)面。將實(shí)驗(yàn)大鼠隨機(jī)分.為空白組、對(duì)照組和實(shí)驗(yàn)組。實(shí)驗(yàn)組創(chuàng)面外.用.益美膚,每日三次;對(duì)照組創(chuàng)面外用比亞芬,每日三次;空白組創(chuàng)面不予處理。于放射后第1、3、5、7、14、21、35天,觀察各組大鼠放射部位皮膚創(chuàng)面損傷及愈合情況后取放射部位皮膚組織,石蠟包埋切片后,一部分用于做HE染色用于組織病理學(xué)檢查,另一部分做免疫組織化學(xué)檢測(cè)(SP法)。使用Ipp6.0圖像軟件檢測(cè)不同時(shí)相點(diǎn)創(chuàng)面表皮干細(xì)胞標(biāo)記物K19和整合素β1陽(yáng)性細(xì)胞表達(dá)情況。所有數(shù)據(jù)用SPSS18.0統(tǒng)計(jì)軟件進(jìn)行單因素方差分析,以p0.05為有統(tǒng)計(jì)學(xué)意義。結(jié)果:1、大體觀察:從照射后的第3天開(kāi)始,空白組照射區(qū)皮膚出現(xiàn)干性脫屑等反應(yīng),甚至出現(xiàn)局部充血性紅斑。對(duì).照組及實(shí)驗(yàn)組在照射后第5天開(kāi)始出現(xiàn)紅斑反應(yīng),但無(wú)明顯脫屑,紅斑顏色較淺;隨著時(shí)間推移,空白組創(chuàng)面顏色加深明顯并形成苔蘚樣壞死潰瘍痂皮,創(chuàng)面脫痂愈合延遲,愈合過(guò)程中逐步變厚變硬呈攣縮狀態(tài),表面不平整,色澤較正常皮膚深,充血水腫明顯,毛發(fā)生長(zhǎng)稀疏。對(duì)照組.實(shí)驗(yàn)組愈合過(guò)程類(lèi)似,被覆新生上皮,愈合部位較平整,顏色與正常皮膚相仿,毛發(fā)生長(zhǎng)相對(duì)較多。2、創(chuàng)面組織學(xué)觀察:與空白組比較,對(duì)照組與實(shí)驗(yàn)組的毛細(xì)血管腫脹程度較輕,到14天后新生毛細(xì)血管逐步增多,上皮細(xì)胞增生較活躍,炎癥反應(yīng)較輕。3、免疫組化結(jié)果:K19及integrinβ1在空白組、對(duì)照組、實(shí)驗(yàn)組均有陽(yáng)性表達(dá),在基底細(xì)胞層及毛囊周?chē)梢?jiàn)大量棕黃色顆粒,其中實(shí)驗(yàn)組陽(yáng)性細(xì)胞數(shù)量最多?.白.組.在.同.一.時(shí).相.點(diǎn).的.K19、β1整.合.素IOD.值.分.別與對(duì)照組、實(shí).驗(yàn).組.比較差異有統(tǒng)計(jì)學(xué)意義(p0.05),對(duì)照組與實(shí)驗(yàn)組比較差異無(wú)統(tǒng)計(jì)學(xué)意義(p0.05)。各組K19、integrinβ1的IOD值均在第21天達(dá)峰值,尤以實(shí)驗(yàn)組峰值最高,隨后表達(dá)逐步降低。結(jié)論:SD大鼠急性β射線Ⅱ度皮膚損傷創(chuàng)面外用益美膚后,表皮干細(xì)胞標(biāo)記物integrinβ1及K19表達(dá)顯著上調(diào),增殖較活躍,加速創(chuàng)面上皮化進(jìn)程,明顯縮短創(chuàng)面愈合時(shí)間。益美膚對(duì)治療大鼠急性β射線Ⅱ度皮膚損傷創(chuàng)面具有確切的療效,療效與比亞芬相當(dāng),具有臨床使用和推廣價(jià)值。
[Abstract]:Objective: to investigate the effect of Yimei skin on repairing the wounds of SD rats with acute 尾 -ray injury by comparing the gross changes, histological changes and the expression of K19 and integrin 尾 _ 1 positive cells in epidermal stem cells.Methods: two to three months old clean grade SDS.102 rats, weighing 190 鹵10 g, were fed alone for 5 days to adapt to the environment.SIMENS linear accelerator was used in the preparation of experimental animal wounds. 6Me V high energy electron beam was set up and exposed skin on the buttocks of SD rats was irradiated for 11 minutes for 52 seconds. The wounds of acute 尾 -ray second-degree skin injury were made in SD rats.The experimental rats were randomly divided.Blank group, control group and experimental group.The experimental group was outside the wound.With.The wounds in the control group were treated with Biafen three times a day, and the wounds in the blank group were not treated.On the 35th day after irradiation, the skin tissue was taken from the radiation site of the rats after the injury and healing of the skin wound. After paraffin embedded sections, some of them were used for HE staining for histopathological examination.The other part was detected by immunohistochemical method.Ipp6.0 image software was used to detect the expression of K19 and integrin 尾 1 positive cells.All the data were analyzed by single factor ANOVA with SPSS18.0 software.Results 1. Gross observation: from the 3rd day after irradiation, dry desquamation appeared in the skin of the blank group, and even local congestive erythema appeared.Yes.The erythema reaction began to appear in the irradiation group and the experimental group on the 5th day after irradiation, but there was no obvious desquamation, and the erythema color was lighter. With the passage of time, the wound color of the blank group deepened obviously and formed the mossy necrotic ulcer callus, and the wound descab healing was delayed.During the healing process, thickening and hardening showed contracture, uneven surface, deeper color than normal skin, obvious hyperemia and edema, and sparse hair growth.Control group.The healing process of the experimental group was similar, the healing site was flat, the color was similar to the normal skin, and the hair growth was relatively more .2.The wound histological observation: compared with the blank group, the healing process of the experimental group was similar to that of the control group.The capillary swelling degree of the control group and the experimental group was mild. After 14 days, the neovascularization gradually increased, the epithelial cells proliferated more actively, and the inflammatory reaction was lighter. The immunohistochemical results showed that: K19 and integrin 尾 1 were in the blank group, and in the control group.A large number of brown granules were found in the basal cell layer and around the hair follicle, among which the number of positive cells in the experimental group was the most.Empty.White.Group.In.The same.I.When.PhasePoint.K19, 尾 1 integral.Together.Simple IOD.Value.Minutes.Do not and control group, solid.Test.Group.The difference was statistically significant (P 0.05), but there was no significant difference between the control group and the experimental group (P 0.05).The IOD value of K19 integrin 尾 1 reached its peak on the 21st day, especially in the experimental group, and then decreased gradually.Conclusion the expression of integrin 尾 1 and K19, the marker of epidermal stem cells, is up-regulated, the proliferation is more active, the process of wound epithelization is accelerated, and the wound healing time is shortened obviously after topical application of the skin in the rats with acute 尾 -ray skin injury.Yimeifu has a definite curative effect on acute 尾 -ray second-degree skin injury wound in rats, and the curative effect is equivalent to that of Biafen, which has clinical application and popularization value.
