【摘要】:目的:探討長(zhǎng)期有氧運(yùn)動(dòng)干預(yù)對(duì)腦衰老過(guò)程中大鼠學(xué)習(xí)記憶能力以及海馬腦源性神經(jīng)營(yíng)養(yǎng)因子(BDNF)表達(dá)的影響。方法:將30只3月齡SD雄性大鼠隨機(jī)分成青年對(duì)照組(YC組)、衰老對(duì)照組(OC組)和衰老運(yùn)動(dòng)組(OS組)3組,每組10只。OC組和OS組腹腔注射D-半乳糖100 mg/kg/d,持續(xù)6周,OS組同期每日進(jìn)行1小時(shí)游泳運(yùn)動(dòng)。6周后用Morris水迷宮實(shí)驗(yàn)評(píng)估各組大鼠的學(xué)習(xí)與記憶能力;然后斷頭取大鼠海馬,分別采用Real Time-PCR和Western Blot技術(shù)檢測(cè)其BDNF m RNA和BDNF蛋白的表達(dá)水平;取大腦皮質(zhì),進(jìn)行自由基指標(biāo)檢測(cè)。結(jié)果:在Morris水迷宮定位航行試驗(yàn)中,與YC組相比,OC組平均逃避潛伏期顯著延長(zhǎng)(P0.01);與OC組相比,OS組平均逃避潛伏期縮短(P0.01)。在空間探索實(shí)驗(yàn)中,OC組平均穿越平臺(tái)區(qū)域次數(shù)顯著性少于YC組與OS組(P0.01,P0.05)。OC組海馬BNDF m RNA和BNDF蛋白表達(dá)水平均低于YC組(P0.01,P0.05),而OS組海馬BNDF m RNA和BNDF蛋白表達(dá)水平高于OC組(P0.01,P0.05)。結(jié)論:衰老過(guò)程中進(jìn)行有氧運(yùn)動(dòng)可延緩腦衰老引起的學(xué)習(xí)記憶能力下降。有氧運(yùn)動(dòng)可上調(diào)衰老過(guò)程中大鼠海馬BDNF的表達(dá),這可能是有氧運(yùn)動(dòng)延緩腦衰老、改善學(xué)習(xí)記憶能力的分子機(jī)制之一。
[Abstract]:Aim: to investigate the effects of long-term aerobic exercise on learning and memory ability and the expression of brain-derived neurotrophic factor (BDNF) in hippocampus during brain aging in rats.Methods: thirty 3-month-old male Sprague-Dawley rats were randomly divided into two groups: the young control group (YC), the aging control group (OC) and the aging exercise group (OS-group).10 rats in each group were injected intraperitoneally with Dgalactose 100 mg / kg / d for 6 weeks. The rats in OS group were subjected to 1 hour swimming for 6 weeks.
