海帶多糖對大鼠頜下腺血管輻射損傷防護作用的初步研究

【摘要】:目的放療為頭頸部腫瘤常用治療方式之一。由于涎腺解剖位置及其周組織緊密毗鄰,在治療過程中輻射損傷無法避免。目前射線引發(fā)腺泡和導(dǎo)管細胞的凋亡為研究熱點,但腺體其他附屬組織如血管、神經(jīng)、淋巴等的損傷亦不容忽視。血管為腺體組織汲取營養(yǎng)及新陳代謝的重要媒介,射線引起腺體血管的損傷及閉鎖可加劇病情惡化。褐藻類萃取物海帶多糖(laminaria japonica polysaccharide, LJP)天然且毒副作用小,具有抗輻射、調(diào)節(jié)免疫、抗氧化等許多生物活性。本研究將探討海帶多糖對60Coγ射線所誘導(dǎo)的大鼠頜下腺血管損傷的防護作用。 方法70只Wistar大鼠隨機分為4組:正常對照組(N組),放療對照組(R組),海帶多糖高劑量(300mg/kg)照射組(LHR組),海帶多糖低劑量(30mg/kg)照射組(LLR組)。照射前3天及照射后3天分別給予腹腔注射生理鹽水和海帶多糖。R組,LHR組,LLR組的大鼠頭頸部予以60Coγ射線15Gy一次性照射。免疫組織化學(xué)方法檢測腺體微血管密度(microvessel density, MVD); RT-PCR檢測血管內(nèi)皮生長因子(vascular endothelial growth factor, VEGF)、基質(zhì)金屬蛋白酶9(matrix metalloproteinase-9,, MMP-9)及基質(zhì)金屬蛋白酶抑制劑1(tissue inhibitor ofmetalloproteinase-1, TIMP-1)的表達,其擴增產(chǎn)物經(jīng)電泳后行凝膠成像分析;透射電鏡觀察頜下腺血管內(nèi)皮細胞超微結(jié)構(gòu)改變。 結(jié)果 ①免疫組織化學(xué)檢測見海帶多糖可降低照射后的頜下腺微血管密度的減小幅度,LHR和LLR組的MVD與R組相比差異有統(tǒng)計學(xué)意義(P0.05); ②RT-PCR檢測VEGF、MMP-9在LHR及LLR組的表達均高于R組(P0.05), TIMP-1在各組表達差異無統(tǒng)計學(xué)意義(P0.05); ③電鏡下觀察R組血管及內(nèi)皮細胞損傷最為明顯,LHR及LLR組損傷較輕。 結(jié)論海帶多糖可減輕射線對頜下腺血管的損傷。
[Abstract]:The purpose of radiotherapy of head and neck cancer is one of the commonly used treatment. Because salivary gland anatomical site and periodontal tissue closely adjacent to the injury can not be avoided in the course of treatment. The radiation ray induce apoptosis of acinar and duct cells as the research hotspot, but the glands and other affiliated organizations such as blood vessels, nerves, lymph and other damage should not be ignored. Vascular glands learn the important nutrition and media organization caused by radiation damage and The new supersedes the old., atresia gland blood vessel can aggravate disease progression. The brown algae extract of Laminaria japonica polysaccharides (Laminaria japonica polysaccharide, LJP) natural and low adverse effect, with anti radiation, regulating immunity, antioxidant and many other biological activities. This study will explore the protective effect of Laminaria japonica polysaccharide induced by 60Co gamma ray submandibular vascular injury in rats.
Methods 70 Wistar rats were randomly divided into 4 groups: normal control group (N group) and radiotherapy group (R group), kelp polysaccharide high dose (300mg/kg) irradiation group (LHR group), low dose kelp polysaccharide (30mg/kg) irradiation group (LLR group). 3 days before irradiation and irradiation after 3 days were given intraperitoneal injection of saline and laminarin.R group, LHR group, LLR group of rat neck to 60Co gamma ray irradiation. 15Gy immunohistochemical method to detect microvessel density (microvessel gland density, MVD RT-PCR); vascular endothelial growth factor (vascular endothelial, growth factor, VEGF), matrix metalloproteinase 9 (matrix, metalloproteinase-9, MMP-9) and tissue inhibitor of matrix metalloproteinase 1 (tissue inhibitor, ofmetalloproteinase-1, TIMP-1) expression of the amplified products were electrophoresis gel imaging analysis; observe vascular endothelial cell ultrastructure by transmission electron microscopy of submandibular gland Structural changes.
Result
(1) immunohistochemistry showed that Laminaria Polysaccharide decreased the microvessel density of submandibular gland after irradiation. There was a significant difference in MVD between LHR and LLR group compared with R group (P0.05).
(2) the expression of VEGF by RT-PCR and the expression of MMP-9 in LHR and LLR groups were higher than that in group R (P0.05), and there was no significant difference in the expression of TIMP-1 in each group (P0.05).
(3) the most obvious damage of vascular and endothelial cells in R group was observed under electron microscope, and the injury of group LHR and LLR was lighter.
Conclusion the polysaccharide of kelp can reduce the damage to the blood vessels of the submandibular gland.
