6周遞增跑臺(tái)運(yùn)動(dòng)對(duì)大鼠腎上腺ACTH、GC受體和血清GC的影響

【摘要】:目的探討6周遞增跑臺(tái)運(yùn)動(dòng)對(duì)大鼠血清GC和腎上腺ACTH、GC受體的影響。方法 SPF級(jí)雄性SD大鼠128只,隨機(jī)分成16組,分別是完全不訓(xùn)練組(A),共4小組,每小組8只;處死當(dāng)天不訓(xùn)練組(B),共4小組,每小組8只;運(yùn)動(dòng)后即刻處死組(C),共4小組,每小組8只;運(yùn)動(dòng)后3 h處死組(D),共4組,每小組8只。A組在整個(gè)實(shí)驗(yàn)過程中不訓(xùn)練,B、C、D組進(jìn)行6周遞增負(fù)荷跑臺(tái)運(yùn)動(dòng)。分別在第0、2、4、6周周末處死A、B、C、D組各一小組。取腎上腺和血液,用Real-time PCR測(cè)試各小組腎上腺ACTH-R mRNA和GR mRNA的表達(dá),用ELISA法測(cè)試各小組血清中GC的含量。結(jié)果第0、2、4、6周末4組大鼠的腎上腺中ACTH-R mRNA在第0周、第2周末和第6周末的表達(dá)沒有顯著性差異(P㧐0.05),只是在第4周末B組和C組比A組和D組有顯著性升高(P0.05);各組GR mRNA在第0周和第2周末無顯著性差異,第4周末和第6周末的C組和D組比其它兩組有極顯著性升高(P0.001);各周末血清中GC含量都表現(xiàn)為C組和D組比其它兩組有顯著性升高(P0.05)。結(jié)論 6周遞增負(fù)荷運(yùn)動(dòng)過程中,腎上腺ATCH-R mRNA表達(dá)表現(xiàn)出時(shí)相性和飽和性,其表達(dá)并沒有和ACTH陽(yáng)性細(xì)胞的表達(dá)保持一致,二者具有不同步性,可能是ACTH在運(yùn)輸或在正、負(fù)反饋調(diào)節(jié)環(huán)節(jié)受到調(diào)控的結(jié)果。隨著運(yùn)動(dòng)強(qiáng)度的增大,機(jī)體的中GR mRNA表達(dá)的含量降低,其表達(dá)趨勢(shì)和GC分泌趨勢(shì)相反,這應(yīng)該是機(jī)體保持自身穩(wěn)態(tài)的保護(hù)機(jī)制之一。
[Abstract]:Objective to investigate the effect of 6-week incremental treadmill exercise on serum GC and adrenal ACTHN GC receptors in rats. Methods SPF grade male SD rats were randomly divided into 16 groups with 8 rats in each group. On the day of death, 4 groups (8 rats per group) were killed without training, 4 groups (8 rats) were killed immediately after exercise, 4 groups (8 rats per group) were killed immediately after exercise, and 4 groups (4 groups) were killed 3 hours after exercise. During the whole experiment, 8 rats in group A were killed at the end of the 6th week, and the adrenal gland and blood were taken from adrenal gland and blood. The expression of ACTH-R mRNA and gr mRNA in adrenal gland was measured by Real-time PCR, and the content of GC in serum was measured by ELISA method. Results ACTH-R mRNA in adrenal gland of 4 groups was detected at the end of the 6th week. There was no significant difference in expression between the 2nd weekend and the 6th weekend. There was no significant difference in gr mRNA between group B and group C at the end of the 4th week compared with that in group A and group D, but there was no significant difference in gr mRNA between group A and group D at the end of week 0 and week 2, but there was no significant difference between group B and group C at the end of the 4th week. At the end of the 4th and 6th weekend, the serum GC levels in group C and group D were significantly higher than those in group C and group D compared with the other two groups. Conclusion during 6 weeks of incremental load exercise, the serum GC levels in group C and group D were significantly higher than those in group C and group D, respectively. The expression of ATCH-R mRNA in adrenal gland showed chronotropic and saturation, and its expression was not consistent with the expression of ACTH positive cells. The expression of ACTH was different from that of ACTH positive cells, and it may be that the expression of ACTH was in transit or positive. With the increase of the intensity of exercise, the expression of gr mRNA in the body decreased, which was contrary to the tendency of GC secretion, which should be one of the protective mechanisms to maintain the homeostasis of the body.
