八珍加肉桂補(bǔ)骨脂湯對運(yùn)動(dòng)疲勞大鼠海馬細(xì)胞凋亡和bcl-2、bax蛋白表達(dá)的影響

【摘要】:[研究目的]:通過對大鼠運(yùn)動(dòng)疲勞模型的建立,從大鼠海馬細(xì)胞凋亡情況以及bcl-2、bax蛋白表達(dá)水平變化的角度來研究八珍加肉桂補(bǔ)骨脂湯對運(yùn)動(dòng)疲勞的保護(hù)作用和具體機(jī)制。[研究方法]:采用SD雄性大鼠45只,體重(185±10)克,根據(jù)實(shí)驗(yàn)要求將大鼠隨機(jī)分為3組:對照組(S,N=15)、模型組(E,N=15)、中藥加運(yùn)動(dòng)組(CE,N=15)。除去對照組不做任何訓(xùn)練之外,其它2組按計(jì)劃進(jìn)行7周逐級增加負(fù)荷的跑臺運(yùn)動(dòng)訓(xùn)練,以此建立慢性運(yùn)動(dòng)性疲勞模型。對照組無任何的運(yùn)動(dòng)訓(xùn)練,正常飼養(yǎng);模型組在每天訓(xùn)練結(jié)束后半小時(shí)之內(nèi),灌胃一次安慰劑生理鹽水;中藥加運(yùn)動(dòng)組訓(xùn)練結(jié)束后半小時(shí)之內(nèi),灌胃一次復(fù)方中藥八珍加肉桂補(bǔ)骨脂湯。[研究結(jié)果]:(1)大鼠海馬組織5-羥色胺、多巴胺含量的比較:和S組比較,E組的大鼠海馬組織5-HT的含量明顯上升,而多巴胺的含量則明顯下降(P0.05);和E組比較,CE組的大鼠海馬組織5-HT的含量明顯下降,而多巴胺的含量明顯上升(P0.05)。(2)大鼠海馬組織細(xì)胞凋亡情況的比較:和S組比較,E組大鼠海馬組織中凋亡的細(xì)胞數(shù)量大大增加(P0.05),具有顯著性差異;和E組比較,CE組大鼠海馬組織中凋亡細(xì)胞的數(shù)量大大減少(P0.05),具有顯著性差異。(3)大鼠海馬細(xì)胞bcl-2蛋白表達(dá)的比較:和S組比較,E組大鼠海馬細(xì)胞內(nèi)bcl-2蛋白表達(dá)水平較明顯上升(P0.05);和E組比較,CE組大鼠海馬細(xì)胞內(nèi)bcl-2蛋白表達(dá)水平明顯上升(P0.05)。(4)大鼠海馬細(xì)胞bax蛋白表達(dá)的比較:和S組比較,E組大鼠海馬細(xì)胞內(nèi)bax蛋白表達(dá)水平明顯上升(P0.05);和E組比較,CE組大鼠海馬細(xì)胞內(nèi)bax蛋白表達(dá)水平明顯下降(P0.05)。[結(jié)論]:(1)八珍加肉桂補(bǔ)骨脂湯通過對多巴胺、5-羥色胺等中樞神經(jīng)遞質(zhì)指標(biāo)的調(diào)節(jié),達(dá)到延緩了運(yùn)動(dòng)性中樞疲勞的發(fā)生。(2)八珍加肉桂補(bǔ)骨脂湯在一定程度上抑制了運(yùn)動(dòng)疲勞大鼠海馬組織內(nèi)的細(xì)胞凋亡,對中樞神經(jīng)細(xì)胞起到了保護(hù)作用。(3)八珍加肉桂補(bǔ)骨脂湯通過對bcl-2和bax蛋白表達(dá)水平的調(diào)節(jié),抑制了細(xì)胞凋亡,對中樞神經(jīng)細(xì)胞起到了保護(hù)作用,從而在一定程度上緩解了運(yùn)動(dòng)性中樞疲勞的發(fā)生。
[Abstract]:[objective]: to establish a model of exercise fatigue in rats. From the point of view of apoptosis of rat hippocampal cells and the change of bcl-2nbax protein expression, the protective effect and specific mechanism of Bazhen plus cinnamon psoralen decoction on exercise fatigue were studied. [methods]: 45 SD male rats, weight of 185 鹵10 g, were used. The rats were randomly divided into three groups according to the requirements of the experiment: control group, model group, Chinese herbal medicine plus exercise group. Except for the control group without any training, the other two groups were trained for 7 weeks in order to increase the load step by step. The model of chronic exercise fatigue was established. The control group had no exercise training and was raised normally. The model group was given a placebo saline once a day within half an hour after the training, and the Chinese medicine plus exercise group was fed within half an hour after the training. The contents of 5-hydroxytryptamine and dopamine in hippocampal tissue of rats were compared: compared with group S, the content of 5-HT in hippocampal tissue of rats in group E was obviously increased, and the content of 5-HT in hippocampus of rats in group E was significantly higher than that in group S, and the content of 5-HT in hippocampus of rats in group E was significantly higher than that in group S. Compared with group E, the content of 5-HT in hippocampal tissue of rats in CE group was significantly decreased, and the content of 5-HT in hippocampus of rats in group E was significantly lower than that in group E, while the content of dopamine decreased significantly. Compared with group S, the number of apoptotic cells in hippocampal tissue of rats in group E was significantly increased, and there was significant difference in the number of apoptotic cells between group E and group S; Compared with group E, the number of apoptotic cells in hippocampal tissue of rats in CE group decreased significantly (P 0.05) and the expression of bcl-2 protein in hippocampal cells of rats was significantly different. Compared with group S, the expression level of bcl-2 protein in hippocampal cells of rats in group E was higher than that in group S. Compared with group E, the expression of bcl-2 protein in hippocampal cells of rats in CE group was significantly higher than that in group E, and the expression of bax protein in hippocampal cells of rats in group E was significantly higher than that in group S. The expression level of bax protein in hippocampal cells of rats in group E was significantly higher than that in group S. Compared with group E, the expression level of bax protein in hippocampal cells of rats in the CE group decreased significantly. [conclusion] Bazhen and Pengguzhi decoction can regulate the central neurotransmitters, such as dopamine 5-hydroxytryptamine, by regulating the expression of P0.05 protein in the hippocampus of the rats in the CE group. [conclusion] Bazhen + Psoralea decoction can regulate the neurotransmitters, such as dopamine serotonin, and so on. Bazhen plus cinnamon psoralen decoction inhibited apoptosis in hippocampus of exercise fatigue rats to some extent. Bazhen plus cinnamon psoralen decoction inhibits apoptosis and protects central nervous cells by regulating the expression of bcl-2 and bax proteins. To a certain extent, it alleviates the occurrence of exercise-induced central fatigue.
